摘  要:[目的]评价多聚赖氨酸修饰的脱钙骨基质支架材料(PLL-DBM)的组织相容性。[方法]采用标准的毒理学方法进行溶血实验、全身急性毒性实验、致癌实验和皮下植入实验验证多聚赖氨酸修饰的脱钙骨基质支架材料(PLL-DBM)的组织相容性。[结果]溶血实验测得溶血指数为2.617%,符合生物材料的国家标准(<5%);全身急性毒性实验:各组裸鼠活动正常,体重正常增长,未见中毒表现或不良反应,72 h内PLL-DBM材料浸提液组与生理盐水组裸鼠体重增长无明显差异(P>0.05),且4周内裸鼠无死亡;致癌实验显示实验组裸鼠正常存活,8个月内未见肿块形成,心、肝、脑、肺、肾、胰、脾组织学观察未见肿瘤细胞;皮下植入实验显示实验动物伤口未见感染、红肿及裂开等不良反应,置入材料无排异现象,取材时见材料周围肌肉颜色及质地正常。[结论]多聚赖氨酸修饰的脱钙骨基质支架材料(PLL-DBM)具有良好的组织相容性。[Objective] To evaluate the histocompatibility of demineralized bone matrix (DBM) decorated with poly -L- lysine(PLL) for scaffolds. [Method] With standard toxicological metheds,the biocompatibility of DBM was evaluated by the he- molysis test, acute systemic toxicity experiment, carcinogenicity study and subcutaneous implant test. [ Result ] The hemolytic in- dex was 2.617% ,and it well met the national standard ( 〈 5% ). No animal died and no toxicity symptom or adverse effects, were noticed within 72 hours. The mean dally weight increase showed no statistically significant difference ( P 〉 0.05 ) between two groups in 72 hours. And no mice died in the next 4 weeks. Carcinngenieity tests showed that the experimental group of mice was normal and no mass formation was observed within 8 months. Histological observation found no tumor cells in heart, liver, brain, lung, kidney, pancreas and spleen. No infection, swelling, cracking of the wounds, or discharge of embedded materials were observed in the subcutaneous implantation experiment. The color and texture of the surrounding muscle were normal when mate- rials were drawn. [ Conclusion] The PLL- DBM has well histocompatibility.
