摘 要:以蛹虫草(Cordycps militaris)子实体为母本,采用组织分离法和子囊单孢子配对法制备子代母种,以母种菌丝形态、平板培养菌丝生长速度、见光培养菌丝转色性能、实际栽培出草结果等为菌种性能主要考察指标,探索优质菌种提纯复壮规律及方法。结果表明,菌种菌丝浓密、健壮、匍匐状,生长快、转色好,则出草形态好、产量高;组织分离法制备母种试管斜面菌丝形态优良率、平板生长及转色优良率、栽培出草优良率可达约95%、90%和85%,分别比子囊单孢子配对法高约10%、25%和40%,后者可产生约20%的更优菌株。说明组织分离法更适合遗传和保持母本性状,子囊单孢子配对法更适合筛选更优菌株,两种方法均可作为蛹虫草优良菌株提纯复壮的方法。The mother plate culture mycelial morphology, mycelial growth rate, light color, the actual performance of mycelia grown grass results of strain performance of Cordyceps militaris were investigated by the quality of C. militaris as female par- ent, tissue isolation method and spore culture method paired preparation of sub mother. And the relevant quality spawn reju- venation rules and method were explored. Results showed that the percentages of fine colonies in tube culture,plate culture, and the percentage of good growth, of spawn prepared by tissue isolation could attain 95% ,90% ,and 85% respectively, which were separately 10%,25% and 40% higher than those spawn prepared by monospore pairing cultivation, although the latter could produce 20% even better colonies. It was suggested that tissue isolation method was more suitable for the conser- vation of genetic and maternal character, while spore culture method was a more appropriate choice for screening better strains,although both methods could be used for the rejuvenation of the fine strain of C. militaris.
