载脂蛋白E基因敲除小鼠血管内皮功能失调模型的建立

摘  要:目的探讨用高脂饮食诱导载脂蛋白E(Apo E)基因敲除小鼠制作血管内皮功能失调模型的可行性。方法高脂饮食饲养Apo E-/-小鼠(Apo E组)以及野生型C57BL/6J小鼠(对照组),8周后,检测两组小鼠血脂、血糖、血胰岛素及一氧化氮水平;取主动脉做病理观察及血管内皮依赖性血管舒张功能实验。结果高脂喂养8周后Apo E组小鼠主动脉出现明显粥样硬化斑块;Apo E组小鼠的血清总胆固醇、HDL-C、LDL-C水平显著高于对照组小鼠(P<0.05),血清一氧化氮水平显著低于对照组(P<0.05);两组小鼠的血糖、血清胰岛素水平均在正常范围,且差异无统计学意义(P>0.05);Apo E组小鼠主动脉内皮依赖性舒张功能最大舒张率为(51.14±6.21)%,显著低于对照组小鼠的(86.52±2.88)%(P<0.05)。结论经过8周高脂饮食喂养的Apo E基因敲除小鼠可以作为研究内皮功能失调的理想动物模型。Objective To explore the feasibility of establishing the vascular endothelial dysfunction model induced by high cholesterol diet in apolipoprotein E(ApoE) knockout mice.Methods ApoE-/-knockout mice(ApoE group) and wild-type(WT) C57BL/6J mice(control group) were fed with high cholesterol diet for 8 weeks.And then the levels of blood lipid,blood glucose,blood insulin and nitric oxide were detected in two groups.The arterial samples were collected for the histopathological observation and endothelium-dependent vasodilatation test. Results Significant aortic atherosclerotic plaques were observed in ApoE-/-mice after 8-week high cholesterol diet.The mice in the ApoE group had significantly higher levels of serum TC,HDL-C and LDL-C as well as significantly lower serum nitric oxide level compared with the controls (P〈0.05),were normal.The maximum arterial endothelium-dependent vasodilatation value of ApoE group was significantly lower than that of control group ((51.14 ±6.21)% vs.(86.52 ±2.88)%,P 〈0.05).Conclusion ApoE knockout mice which have been fed with high cholesterol diet for 8 weeks can act as an ideal animal model for endothelial dysfunction research.
