摘 要:以催乳素受体基因(prolactin receptor,PRLR)作为影响鸡产蛋性能的候选基因,采用连接酶检测反应(ligase detection reaction,LDR)对378只四明香鸡B系的PRLR基因进行多态性检测,并利用最小二乘法分析了4个突变位点SNP1(C11390G),SNP2(A14952G),SNP3(A14969C)和SNP4(A14984G)与鸡134日龄体重、65周体重、65周产蛋数和300日龄产蛋数的相关性。单位点分析结果显示SNP2/SNP3/SNP4完全连锁,且其位点多态性对300日龄产蛋数影响显著(P<0.05),G/A/G基因型300日龄产蛋数显著高于A/C/A基因型7枚(P<0.05);SNP1位点多态性与鸡134日龄体重、65周体重、300日龄产蛋数和65周产蛋数均无显著关联(P>0.05)。单倍型分析结果发现存在3种单倍型,即H1(CACA),H2(GACA)和H3(CGAG),不同单倍型对300日龄产蛋数差异显著(P<0.05),表现出H3(CGAG)>H2(GACA)>H1(CACA)的趋势,且H3(CGAG)基因型的300日龄产蛋数比H1(CACA)基因型高7.33枚(P<0.05)。结果表明,PRLR基因中H3(CGAG)为有利单倍型,可作为提高鸡群产蛋性能潜在的分子标记。Prolactin receptor (PRLR) gene was selected as a candidate gene for chicken laying performances in this study. Ligase detection reaction was used to detect 4 polymorphic sites in PRLR gene of 378 chickens of Siming Xiang chicken B strain, namely SNP1 (Cl1390G), SNP2 (A14952G), SNP3 (A14969C) and SNP4 (A14984G). Associations between these SNP polymorphisms and egg production within 65 weeks and 300 d, body weights at 134 d and 65 weeks, were analyzed by the least square analysis. As for single site analysis, results showed that SNP2/ SNP3/SNP4 was significantly ( P 〈 0. 05 ) associated with egg production within 300 d, and egg production within300 d of genotype G/A/G was significantly ( P 〈 0. 05 ) higher than that of A/C/A with 7 eggs, while SNP1 (C11390G) had no significance on the all traits detected. As for haplotype analysis, results showed that 3 haplotype for 4 SNPs of PRLR gene was detected, which were H1 (CACA), H2 (GACA) and H3 (CGAG), and different hap- lotype had significant (P 〈 0. 05) differences on egg production within 300 d. The predominant haplotype was H3 (CGAG) 〉 H2(GACA) 〉 H1 (CACA) successively, in which hens of haplotype H3 (CGAG) had 7.33 eggs more than that of H1 (CACA). It indicated that H3 (CGAG) of chicken PRLR gene was the best haplotype for egg produc- tion, which was the potential marker for laying performance.
