摘  要:为探讨辐射对离体培养的甘蓝型油菜再生的影响,试验选择再生频率较高的甘蓝型黄籽油菜品系SH31、K81和甘蓝型黑籽品系P52,以种子、预培养的下胚轴为诱变材料进行不同剂量的^60Co-γ射线照射处理,分析了辐射对油菜芽再生的影响。结果显示:甘蓝型油菜种子经^60Co-γ射线辐射后离体培养,以下胚轴为外植体,外植体芽再生率随^60Co-γ剂量的增加而降低;用不同剂量的γ射线直接对预培养的下胚轴进行辐射,10~30 Gyγ射线辐射可明显促进外植体的再生,并且随浓度增加而增加,但是当浓度超过40 Gy,再生率则随浓度增加而降低。另外预培养的时间对辐射后芽的再生影响较大,预培养4 d的外植体芽再生率高于预培养2 d的。Effects of irradiation treatment on regeneration freqency of hypocotyls in Rape B. napus were studied so as to select the two yellow-seeded rapeseed lines( SH31, K81) with high regeneration frequency and one black-seeded rapeseed line P52 as the main experimental materials to study the related effects in rape after oilseeds, pre-culture hypocotyls, were treated with γ irradiation. The regeneration rate of pre-culture hypocotyls explants from rape(B. napus L) seeds irradiated by ^60Co-γ rays decreased with the increase of irradiation dose. Induction frequency of adventitious buds increased by irradiation treatment of γ-ray with low dose of 10~30 Gy to explants in pre-culture, but decreased when the dose was higher than 40 Gy. In addition, the pre-culture time influenced the ratio of regeneration. The regeneration frequency of hypocotyls by 4-day pre-culture was higher than by 2-day pre-culture.
