广西H3N8亚型马流感病毒耐药性分析

摘　　要：【目的】了解广西H3N8亚型马流感病毒(Equine influenza virus,EIV)对抗流感病毒药物的耐药性,为临床科学用药提供参考。【方法】采用RT-PCR对广西H3N8亚型EIV分离株A/Equine/Guangxi/1/09(简称GX09株)进行M基因和NA基因扩增,测序分析M2基因和NA基因特征及其与耐药性相关的氨基酸位点,并进行生物学耐药性试验。【结果】GX09株M2基因的相关耐药性位点(L26F、V27A、A30T、S31N、G34E)均未发生变异;在NA基因相关耐药性位点方面,也仅在S247A位点处出现氨基酸替换,其他位点均较保守,未出现氨基酸替换。生物学耐药性试验结果表明,GX09株对金刚烷胺敏感,对奥司他韦已产生耐药性。【结论】广西目前流行的EIV对金刚烷胺敏感,在流感易发季节仍可选用烷胺类药物对广西地区的马流感进行预防和治疗。【Objective】The present experiment was conducted to investigate anti-influenza drug resistance of equine influenza virus（EIV） isolated from Guangxi to provide references for scientific clinical medication. 【Methods】The M and NA gene fragments of were amplified by RT-PCR from EIV subtype H3N8 strain A/Equine/Guangxi/1/09（GX09 for short） isolated from Guangxi. Then, the genetic traits of M2 and NA and its amino acid loci related to drug resistance were analyzed after sequencing. And the drug-resistance of obtained isolates was also assayed by biological method. 【Results】The results showed that there was not mutational amino acid among all amino acids（L26F, V27 A, A30 T, S31 N, G34E） related to drug resistance of M2 protein. And there was not amino acid to be substituted among all amino acids related to drug resistance of NA protein, except S247 A mutation. The result of biological resistance experiments indicated that GX09 strain was sensitive to amantadine, however resistant to oseltamivir. 【Conclusion】The epidemic EIV（GX09 strain） of Guangxi was also sensitive to amantadines. Therefore amantadine should be used to prevent and treat EIV of Guangxi in flu outbreaks season.
