HPLC测定母草属药用植物中的齐墩果酸和绿原酸

摘　　要：目的采用HPLC法测定母草属药用植物中的齐墩果酸和绿原酸。方法采用Agilent TC-C18色谱柱(250 mm×4.6mm,5μm),齐墩果酸的流动相为乙腈-0.5%乙酸铵(85∶15),流速0.8 m L·min-1,检测波长205 nm;绿原酸的流动相为乙腈-0.4%磷酸并进行梯度洗脱,流速1 m L·min-1,检测波长327 nm。结果标准曲线方程分别为:Y齐墩果酸=2.8737×105X-1.2185×105(r=0.9998)、Y绿原酸=9.5946×105X-1.5664×105(r=0.9999),线性关系良好;7种药用植物中母草、狭叶母草、长蒴母草含齐墩果酸和绿原酸,其中,长蒴母草中齐墩果酸的含量超过0.2%,绿原酸的超过0.5%;宽叶母草、陌上菜、泥花草、旱田草均含有齐墩果酸,但不含绿原酸。结论长蒴母草中齐墩果酸和绿原酸的含量均较高,具有潜在的开发价值。OBJECTIVE To determine oleanolic acid and chlorogenic acid in medicinal Lindernia All. by HPLC. METHODS An Agilent TC -C18 （250 mm ×4.6 mm,5 μm）was used. The content of oleanolie acid was determined with the mobile phase was a mixture of acetonitrile -0.5 % ammonium acetate （85 ： 15 ） , flow rate was 0.8 mL· min^- 1, and the detection wavelength was 205 nm. The content of chlorogenic acid was determined with the mobile phase of aeetonitrile - 0.4% phosphoric acid in gradient elution, and flow rate of 1 mL· min^- 1, , and the detection wavelength was set at 205 nm. RESULTS The standard curve equation was Yoleanolic acid = 2. 8737 × 10^5X - 1. 2185 × 10^5 （ r = 0. 9998 ） , Ychlorogenio acid = 9. 5946 × 10^5X - 1. 5664 × 10^5 （ r = 0. 9999 ）, and the linear relationship was good. Oleanolic acid and chlorogenic acid was contained in L. crustacea,L, angustifolia and L. anagallis, among which the content of oleanolic acid in L. anagallis was more than 0.2% and the chlorogenic acid more than 0.5%. L. nummularifolia, oleanolic acid was contained in L. procumbens ,L. antipoda and L. ruellioides ,but no chlorogenic acid. CONCLUSION As the high content of oleanolic acid and chlorogenic acid, L. anagallis has developmental values.
