摘 要:目的探讨蝙蝠葛活性成分在体内外抗肿瘤的作用。方法将30只BALB/c A裸小鼠按随机数字表法随机分为阴性对照组、阳性对照组、药物低剂量组、药物高剂量组。进行体外细胞培养,在细胞复苏和培养之后,进行细胞活力检测。使用MTT比色法按照实验标准方法进行。每周对所有小鼠测量移植瘤的长度、宽度,动态观察裸鼠移植瘤的生长情况,并计算体积,制作裸鼠移植瘤的生长去曲线图。结果蝙蝠葛活性成分对人肺癌A-549细胞株,人肝癌Hep G2细胞株,人肝癌Hep3B细胞株,人宫颈癌Hela细胞株、人乳腺癌MCF-7细胞株、人白血病K-562细胞株、人胃癌MGC-803细胞株、人胃癌SGC-7901细胞株和小鼠黑色素瘤B-16细胞株均有明显抑制增殖作用。药物低剂量组的T/C值为68.4%,对裸小鼠移植瘤的生长无明显地抑制作用;药物高剂量组的T/C值为58.8%(P<0.05)。结论蝙蝠葛活性成分在体外具有较强的广谱抗肿瘤细胞增殖作用,且高剂量组蝙蝠葛对移植肿瘤的抑制效果非常明显。Objective: To investigate the anti-tumor effects in vitro and vivo of active component in dauricine. Methods: 30 BALB/cA nude mice were divided into the negative control group,the positive control group,the low-dose group of the active component and the high-dose group of active component cells were cuhured in vitro. After cell recovery and culture,cell viability was tested. MTr colorimetric test was conducted in accordance with standard procedures. Weekly, the length and width of the transplanted tumor were measured. The growth in nude mice was dynamically observed,and the volume and growth were calculated in nude mice to make graphs. Re- suits: Active component of dauricine had an inhibitory effect on human cancers, such as cell line A-549 in lung cancer,cell line HepG2 and Hep3B in hepatoma,cell line Hela in cervical cancer,eel1 line MCF-7 in breast can- cer,cell line K-562 in leukemia,cell line MGC-803 and SGC-7901 in gastric cancer as well as inhibited prolif- eration on cell line B-16 in mouse melanoma. In the low dose group, T/C value was 68.4% with no significant in- hibitory effect;in the low dose group,T/C value was 58.8% (P 〈 0.05). Conclusion: Active component of dauricine in vitro has a strong broad-spectrum anti-tumor cell proliferation ,and dauricine has an obvious in- hibitory effect on transplanted tumor in the high-dose group.
