摘  要:目的繁育和鉴定Notch 1基因敲除小鼠。方法用B6.Notch 1flox/flox和B6.Alb-Cre转基因小鼠进行饲养和杂交;将繁育出的第1代小鼠再进行相互交配,得到第2代小鼠。通过PCR鉴定出基因型为B6.Notch 1flox/flox.Alb-Cre的第2代小鼠。采用Western blot法检测Notch 1蛋白在肝脏中有无表达。结果两种转基因小鼠繁殖产生的第2代小鼠基因型符合孟德尔遗传定律,可通过该方法获得较多的B6.Notch 1flox/flox.Alb-Cre小鼠。该基因型小鼠肝脏中不表达Notch 1蛋白。结论应用Cre-loxp系统可以成功地敲除小鼠Notch 1基因。Objective To identify and breed Notch 1 gene-knockouted mice. Methods B6. Notchch 1flox/flox and B6. Alb-Cre transgenic mice were bred and hybridized. The first generation of offsprings was mated with each other and get the second generation. The genotype of B6. Notch 1flox/flox Alb-Cre mouse of the second generation was identified by PCR. The expression of Notch 1 protein in the liver was detected by Western blot for testing whether Notch 1 gene had been knoekouted. Results The genotype of second-generation offspring was in according with Mendelian laws. More B6. Notch 1flox/flox . Alb-Cre mice could be obtained through this method. No Notch 1 protein was detected in the liver of the second-generation offspring. Conclusion Cre-loxp system could successfully knockout the Notch 1 gene in mice.
