摘 要:目的探讨唾液乳杆菌对哮喘Balb/c小鼠血清及肺组织匀浆中IgE、嗜酸性粒细胞阳离子蛋白(ECP)、转化生长因子(TGF)-B的影响。方法选择30只4周龄、体重16—18g、SPF级Balb/c雌性小鼠,随机分成3组:正常对照组(A组)、哮喘组(B组)、哮喘+唾液乳杆菌组(C组),应用卵蛋白建立急性哮喘模型,观察小鼠行为学改变,病理HE染色观察气道炎症改变,电镜观察气管纤毛数量变化,ELISA法测定血清及肺组织匀浆中ECP、IgE、TGF-β含量。结果(1)B组小鼠在4%卵蛋白生理盐水溶液雾化激发5—10min后出现明显的烦躁不安、头面部瘙痒、抓耳挠腮、呼吸急促、打喷嚏、弓背、前肢缩抬、二便失禁等哮喘急性相表现,C组小鼠以上反应减轻;(2)肺组织病理HE染色见B组支气管管壁增厚、管腔狭窄、气管及血管周围大量以嗜酸性粒细胞为主的炎性细胞浸润,管腔中较多炎性分泌物,C组小鼠肺组织病理改变较B组明显减轻;电镜下,气管纤毛数量B组较A组显著减少,C组较B组显著增加;(3)C组血清IgE[(0.86±0.14)μg/ml]及ECP[(2.30±0.15)ng/ml]表达水平分别较B组血清IgE[(1.30±0.12)μg/ml]及ECP[(3.41±0.15)ng/ml]明显降低(P〈0.05);C组肺组织匀浆IgE[(0.97±0.06)μg/ml]及ECP[(5.88±0.13)ng/ml]含量分别较B组肺组织匀浆IgE[(1.13±0.05)μg/ml]及ECP[(8.08±0.15)ng/ml]明显降低(P〈0.05);C组血清TGF—β[(0.17±0.01)μg/ml]及肺组织匀浆中TGF—p[(0.18±0.01)μg/ml]表达水平分别高于B组血清TGF-B[(0.13±0.01)μg/ml]及肺组织匀浆中TGF-β[(0.15±0.01)μg/ml],差异有统计学意义(P〈0.05)。结论致敏前唾液乳杆菌灌胃减轻了哮喘小鼠的临床症状,一定程度上减轻了哮喘模型小鼠的气道炎症,TGF—β可能参与了唾液乳杆菌发挥保护作用�Objective To explore the influence of Lactobacillus salivarius on IgE,cosinophil cationic protein(ECP) and transforming growth factor(TGF)-β of serum and lung tissue homogenates of asthmic Balb/c mouse. Methods Thirty female Balb/c mice weighing 16-18 g(4 weeks old) ,SPF grade,were randomly divided into three groups:normal control group (group A), asthma group (group B ), asthma combined Lactobacillus salivarius group (group C). Acute asthma model was established applying ovalbumin, behavioral changes were observed in mice, the changes in airway inflammation and the number changes of tracheal cilia were observed by pathological HE staining and the electron microscope respectively. The levels of IgE, ECP and TGF-β in the serum and lung tissue homogenate were measured by ELISA method. Results ( 1 ) The mice in group B presented obvious restlessness, head and facial itching, scratching, shortness of breath, perioral cyanosis with abdominal breathing, sneezing, stooping, forelimbs shrinkage, urine and feces incontinence and other asthma acute phase performances in 5-10 minuts after 4% ovalbumin saline solution atomization excitation,the above reaction alleviated in group C compared to group B. (2) Pathology of HE staining in group B showed the thickening bronchial wall, narrow lumen, peribronchial and perivascular infiltration with a large number of eosinophil-based inflammatory cells, lumen with many inflammatory secretions, pathological changes in lung tissue of mice in group C significantly alleviated compared with group B. Under the electron microscope, the number of cilia in group B significantly decreased compared with group A, while group C significantly increased compared with group B. (3) Expression levels of serum IgE and ECP in group C were significantly lower than those in group B[ (0.86 s0. 14)μg/ml vs. (1.30 s0. 12) μg/ml; (2. 30 ±0. 15) ng/ml vs. ( 3.41 ± 0. 15 ) ng/ml, P 〈 0. 05, respectively ].
