摘 要:目的通过在线生物程序分析刚地弓形虫(Toxoplasma gondii)棒状体蛋白12(ROP12)的生物信息学特性。方法通过在线ProtParam、SOSUI、SOPMA、TMHMM、MotifScan及SYFPEITHI等程序,同时结合DNASTAR、DNAMAN生物信息学软件,分析TgROP12蛋白的生物学特性,如理化性质、二级结构、结构域及抗原表位等。结果TgROP12蛋白含有236个氨基酸,其为分子式C1125H1786N308O360S3,其分子质量和理论等电点分别为25.48ku和4.53。TgROP12蛋白具有信号肽序列和跨膜结构区域,其二级结构中β-转角和无规则卷曲分别占7.20%和44.92%;TgROP12蛋白含有8个亲水性较高(临界值为2.0)的区域,10个表面可及性较高(临界值为1.9)的区域;TgROP12蛋白含有保守结构区域和翻译后修饰位点各6个,并含有9个B细胞抗原表位和6个T细胞抗原表位。结论生物信息学分析显示TgROP12蛋白的免疫原性较好,可作为弓形虫病疫苗候选分子。Objective The bioinformatic characteristics of the rhoptry protein 12 of Toxoplasma gondii were analyzed using bioanalytical software online. This work should lay the foundation for future research into the ROP12 gene. Meth- ods The biological features of the TgROP12 protein including, its physical and chemical properties, secondary structure, domains, and epitopes, were analyzed using software online such as ProtParam, SOSUI, SOPMA, TMHMM, MotifS- can, SYFPEITHI, DNAMAN, and DNASTAR. Results The TgROP12 protein consists of 236 amino acids and has the following characteristics: a molecular formula of C^12~ H1786 N308 0360 $3 ~ a molecular mass of 25.48 ku; a theoretical i- soelectric point of 4.53~ and signal peptide and transmembrane regions. Beta turns accounted for 7.20~ of the secondary structure of the TgROP12 protein while random coils accounted for 44.92~~. There were 8 regions with a high level of hydrophilicity (critical value.. 2.0) and 10 regions with a high level of surface accessibility (critical value.. 1.9). The TgROP12 protein contains 6 conserved regions and 6 post-translational modification sites. The TgROP12 protein also contains 9 B cell epitopes and 6 T cell epitopes. Conclusion The TgROP12 protein has good immunogenicity and may be a candidate for potential vaccines.
