摘 要:目的研究不同浓度的一氧化氮(NO)对库存悬浮红细胞变形性的影响。方法取10例新鲜悬浮红细胞,在储存的不同时相(3 h、1 d、3 d、5 d、7 d、14 d、21 d、28 d),每例和不同浓度L-精氨酸(NO前体)混合培养,浓度分别为1μM(B)、5Μm(C)、10Μm(D)、50Μm(E)。培养后测定红细胞变形性,用NO荧光探针测定反应红细胞内的一氧化氮水平的平均荧光强度(PKPos X)。结果保存期内库存红细胞变形指数、聚集指数和刚性指数均随着时间延长而逐渐增高。当NO前体浓度为5μM时能够明显改善保存14 d内的库存血红细胞变形性指数;高于相应的最佳NO浓度范围的时红细胞变形指数受损。结论 NO对库血红细胞具有调控作用,通过补充最佳浓度的NO(如10μM)时,会使红细胞变形性增加;储存期间红细胞的损伤与NO流失有关,适当补充相应浓度NO可以改善红细胞的变形性。Objective The study was conducted to explore the influence of different concentrations of nitric oxide(NO) on deformability of red blood cell(RBC) suspension stocks. Methods Ten stocks of fresh RBC suspension were cultured with 1 μM(B),5 mΜ(C), 10 mΜ(D), and 50 mΜ(E) of L-arginine(NO precursor) at different time points(3h, 1d, 3d, 5d, 7d, 14 d, 21 d, and28d). RBC deformability was measured after the incubations. The average fluorescence intensity(PKPos X) of NO inside of the RBCs was determined by NO fluorescence probes. Results Erythrocyte deformability index, aggregation index, and rigidity index of the RBC stocks gradually increased with time during storage. Erythrocyte deformability index of the RBC stocks stored for less than 14 d showed significant improvement when incubated with 5 μM of NO precursor. Erythrocyte deformability index began to show reduction when NO concentration is higher than the optimal range. Conclusion NO has regulatory effect on RBC stocks. Erythrocyte deformability would increase when supplemented with the optimal NO concentration(10 μM). RBC damage during storage is associated with NO loss; therefore, supplementation of proper concentrations of NO may improve deformability of erythrocytes.
