摘　　要：目的以脑出血患者血肿周围组织为研究对象,探讨脑出血血肿周围组织IL-17与NF-κBp65的表达及意义。方法选取脑出血行血肿清除术的患者脑组织48例,按发病距手术取材时间分为4组,分别为≤6h组、6-24h组、24-72h组和>72h组,每组12例标本。另选取手术入路中远离血肿的脑组织6例作为对照组。RT-PCR法检测IL-17mRNA的表达,Western blot法检测NF-κBp65核蛋白的表达。结果≤6h组IL-17mRNA的表达与对照组相比无统计学差异(P>0.05),其他各实验组IL-17mRNA的表达与对照组相比有统计学差异(P<0.05)。24-72h组血肿周围组织IL-17mRNA的表达显著高于其他各实验组(P<0.05)。各实验组血肿周围组织NF-κBp65核蛋白的水平与对照组相比均具有统计学差异(P<0.05)。24-72h组血肿周围组织NF-κBp65核蛋白的水平显著高于其他各实验组(P<0.05)。结论脑出血发病6h后,血肿周围脑组织IL-17的表达增高;脑出血发病6h内,脑出血血肿周围脑组织NF-κBp65的表达增高。脑出血后,IL-17/NF-κBp65通路被激活,与脑出血后组织的炎症反应及组织损伤有关。Objective human perihematoma tissue in intracerebral hemorrhage were used to investigate the expres-sion and significance of IL-17 and NF-κBp65.Methods 48 patients with intracerebral hemorrhage who carry out crani-otomy and hematoma removal were selected.Each group were 12 patients.According to the phase between onset to op-eration,All Specimens were divided into the following four groups：≤6 h group,6-24 h group,24-72 h group and〉72 h group.Perihematoma tissue far away from the hematoma was taken as control group.Expression of IL-17 mRNA were detected by RT-PCR.Levels of NF-κBp65 was detected byWestern blot.Results Expression of IL-17 mRNA in ≤6 h group compared with control group had no statistically significant difference （P 〉0.05）,Expression of IL-17 mRNA in other experimental groups had statistically significant difference compared with control group （P 〈0.05）.Expression of IL-17 mRNA in 24-72 h group was significantly higher than that in other groups （P 〈0.05）.the level of NF-κBp65 in experimental groups had statistically significant difference compared with control group （P 〈0.05）.The Level of NF-κBp65 in 24-72h group was significantly higher than that in other experimental groups（P 〈0.05）.Conclusion Expres-sion of IL-17 in perihematoma tissue increased after 6 h,Level of NF-κBp65 in perihematoma tissue increased within 6 h.IL-17/NF-κB pathway was activated after intracerebral hemorrhage.,which was involved in the inflammation and damage in perihematoma tissue.
