摘 要:目的采用Balb/c小鼠成纤维细胞3T3建立新药体外3T3细胞中性红光毒性试验方法,并验证该方法的可靠性。方法利用盐酸氯丙嗪作为阳性药物,建立体外3T3细胞中性红光毒性试验方法 ;然后基于上述试验方法检测6个已知光毒性物质的光毒性,对方法的灵敏度和特异性进行验证。结果在本试验条件下,盐酸胺碘酮、诺氟沙星、蒽和原卟啉均表现出潜在光毒性;而六氯酚和L-组氨酸均无光毒性,试验结果与相关文献报道一致。结论在本试验条件下,本研究成功建立并验证了体外3T3细胞中性红光毒性试验方法,可用于新药的光毒性评价。Objective To establish in vitro 3T3 neutral red uptake phototoxicity test by using Balb/c 3T3 cells for new drug research and to validate its reliability. Methods Chlorpromazine hydrochlorid was selected as the positive control to establish the method of in vitro 3T3 neutral red uptake phototoxicity test. Then the phototoxicity of six known drugs was tested to verify their sensitivity and specificity. Results Under the test conditions, amiodarone hydrochloride, norfloxacin, anthracene and protoporphyrin were considered to be positive in 3T3 neutral red uptake phototoxicity test, while hexachlorophene and L-histidine were negative. The results were consistent with the published data of these drugs. Conclusion Under the test conditions, the in vitro 3T3 neutral red uptake phototoxicity test is successfully established and validated. It can be used to assess the phototoxic effects of new pharmaceuticals.
