摘 要:目的:观察丹参多酚酸盐对心肌梗死后大鼠血管新生和血管生长因子(VEGF)、微小RNA-126(miR-126)表达的影响。方法:将55只雄性SD大鼠随机分为丹参多酚酸盐低剂量组(LS组,n=15)、丹参多酚酸盐高剂量组(HS组,n=15)、模型组(M组,n=15)及假手术组(S组,n=10)。HS、LS及M组大鼠行冠状动脉左前降支结扎术建立急性心肌梗死(AMI)模型后,分别予腹腔注射丹参多酚酸盐30、15 mg/(kg·d)及相应剂量生理盐水。S组只穿线不结扎,予腹腔注射相应剂量生理盐水,每日1次,共7 d。4周后,采用免疫组织化学法检测各组大鼠心肌梗死边缘区微血管密度(MVD)和VEGF的表达;采用实时荧光定量逆转录聚合酶链反应(RT-PCR)法检测心肌梗死边缘区miR-126、VEGF mRNA表达水平。结果:M组MVD、VEGF蛋白以及mRNA表达均高于S组(P<0.05),M组miR-126表达低于S组(P<0.05);LS和HS组MVD、VEGF及miR-126表达均高于M组(P<0.01);HS组MVD、VEGF蛋白、VEGF mRNA及miR-126表达高于LS组(P<0.01)。结论:丹参多酚酸盐对心肌梗死后大鼠血管新生有促进作用,其作用机制可能与上调心肌miR-126表达、促进VEGF表达有关,且呈剂量依赖性。Objective: To investigate the effects of salvianolate on angiogenesis and expression of vascular endothelial growth factor (VEGF) and microRNA-126 ( miR-126 ) after acute myocardial infarction in rats. Methods:A total of 55 male SD rats were randomly allocated to the model(M) group (n = 15), low-dose salvlanolate-treated (LS) group(n = 15), high-dose salvlanolate-treated (HS) group (n = 15)and the sham operated (S)group(n = 10). Left anterior descending coronary artery(LAD) of rats in the M, LS and HS group was ligated to create the models of acute myocardial infarction and rats were given salvlanolate at 15 mg/(kg d),30 mg/(kg d) ,normal saline by intraperitoneal injection for 7 days. The S group was established and given normal saline for 7 days. After 4 weeks, the myocardial microvessel density (MVD) and VEGF in marginal zone of infracted region were detected by inmmnohistochemical staining, and the expression levels of miR-126 and VEGF mRNA were determined by real time RT-PCR. Results: Compared with the S group, the MVD, VEGF protein and VEGF mRNA were higher but the expression of miR-126 was lower in the M group (P〈0. 05), Compared with the M group, the MVD, VEGF protein, VEGF mRNA and miR-126 expression in the LS and HS group all obviously elevated(P〈0. 05). The MVD, VEGF protein, VEGF mRNA and miR-126 expression in the HS group were higher than those in the LS group(P〈0. 05). Conclusion:Salvianolate plays a role in promoting angiogenesis after myocardial infarction, through upregulating the expression of miR-126, VEGF and VEGF mRNA in the ischemic region in a dose dependent manner.
