暑热宁口服液中挥发油含测影响因素研究

摘　　要：目的：考察暑热宁口服液中挥发油含测影响因素,为工艺制定、样品制备、质量标准制定、服药方式等提供依据。方法：以百秋李醇、麝香草酚-香荆芥酚为含测指标,考察过滤方式、提取方式、提取时间、正己烷量、计算方法、p H值、盐析、灭菌、药液、稀释等因素对含测值的影响。结果：过滤和调p H值后各含测指标均降低,提示工艺制定时不可过滤和调节p H值;先超声再提油含测值增大约5%,提示服药前应先温水加热混匀;样品制备宜用成品提油,加4 m L正己烷,提取2.5 h;内标法与外标法含测结果一致;灭菌跟药液本身可使百秋李醇减少约85%,麝香草酚-香荆芥酚减少约21%~24%。结论：诸多因素可以影响挥发油含测值,在进行工艺、质量标准制定和用药指导时要综合考虑。Objective：It was to investigate the affecting measurement factors in the volatile oil containing of Shure＇ning oral liq- uid, providing the basis for technology development, sample preparation, quality standard and the way of medieine. Methods：The patchouli alcohol and thymol - oxyzymol was the measurement indicators, we investigated the influences of filtering methods, ex- traction mode, extraction time, N - hexane quantity, ealeulation method, pH value, salting out, sterilization, liquid and dilution on measured values. Results：After filtration and adjusting pH value, the containing indexes decreased, prompting no filtering or adjus- ting pH value during the process. Ultrasonic before oil extraction made the measured values increase by about 5% ,prompting be- fore taking the medicine it should be heated by warmed water and mixed;sample preparation should use finished oil extraction, adding 4 mL n - hexane, extracting for 2. 5 h. Internal standard method and external standard method had the same measured re- sults consistently. Sterilization and liquid itself can make the patchouli alcohol reduce around 85%, thymol -oxyzymol decrease a- bout 21% -24%. Conclusion：Many factors can affect the volatile oil measured values. In the process, quality standards and guide- lines should be considered.
