金银花水提物体外抗病毒实验研究

摘　　要：目的：为建立金银花抗病毒谱-效相关评价系统,对金银花水提物体外抗病毒活性进行筛选,最后选择金银花水提物抗甲型流感病毒H1N1活性进行研究。方法：采用体外抗病毒实验CPE法结合中性红染色,以半数治疗浓度（EC50）和半数细胞毒性浓度（TC50）并以治疗指数TI作为评价指标,观察金银花水提物对单纯疱疹病毒Ⅰ型、手足口病病毒EV71株和流感病毒H1N1株的抑制作用。结果：金银花水提物对单纯疱疹病毒Ⅰ型、手足口病病毒EV71株未见明显抗病毒效果,对甲型流感病毒H1N1株有显著抗病毒效果且测得80批金银花水提物抗甲型流感病毒H1N1株的治疗指数为14.57~39.85,阳性对照药利巴韦林治疗指数为16.78。结论：金银花具有良好的抗流感病毒H1N1的作用,且某些样品作用优于利巴韦林对照组,实验重复性好,数据真实可靠,为进一步建立金银花谱-效模型提供了科学依据和数据支持。Objective ： To establish the spectrum - efficiency - related evaluation system of Lonicera japonica Thunb, we studied the antiviral effects of Lonicerajaponica Thunb water extract in vitro and selected influenza A virus H1N~ strain（ Flu -A -Hj Nj ） as the suitable virus. Methods ： Adopt anti - virus CPE method combined with neutral red staining to get half therapeutic concentra- tions （ECso） and half of cytotoxic concentration（ TCs0 ） and using therapeutic index（TI） as evaluation index to investigate Lonicera japonica Thunb＇s inhibitory effect on influenza A virus （ Flu - A - H1N1 ）, herpes simplex virus type I （ HSV - 1 ） and Human enterovirus 71 （EV71）. Results：The Lonicerajaponica Thunb water extract showed no antiviral effect on HSV -1 and EV71 strains but had significant antiviral effect on influenza virus. The data of TI of 80 batches of Lonicera japonica Thunb anti - influenza ther-apeutic index of Flu- A- Hi NI was 14.57 -39.85 and the positive control drug ribavirin index was 16.78. Conclusion ：Lonicera japonica Thunb has good effect on anti - Flu - A - H1N1 and some samples are superior to ribavirin control group. The method has good repeatability and the data is true and reliable which provides a scientific basis and data support for the further establishment of Lonicera japonica Thunb spectrum - efficiency model.
