健脾化痰汤对胰岛素抵抗3T3-L1脂肪细胞炎症细胞因子分泌的影响

摘　　要：目的:观察健脾化痰汤含药血清对胰岛素抵抗3T3-L1脂肪细胞分泌脂联素、TNF-α和IL-6的影响,探讨其改善IR可能机制。方法:采用细胞培养技术,将3T3-L1小鼠前脂肪细胞诱导分化为成熟脂肪细胞,通过地塞米松诱导建立IR细胞模型,将脂肪细胞分为正常组(基础培养液加10%空白组血清培养正常脂肪细胞)、IR组(基础培养液加10%空白组血清培养IR脂肪细胞)、中药组(基础培养液加10%健脾化痰组血清培养IR脂肪细胞)和西药组(基础培养液加10%比格列酮组血清培养IR脂肪细胞),培养48 h,葡萄糖氧化酶法检测细胞培养上清液中葡萄糖浓度,ELISA法检测细胞培养上清液中脂联素、TNF-α和IL-6含量。结果:与正常组比较,IR组葡萄糖消耗量和脂联素含量显著降低(P<0.05),TNF-α和IL-6含量显著升高(P<0.05);与IR组比较,中药组和西药组葡萄糖消耗量和脂联素含量明显升高(P<0.05),TNF-α和IL-6含量显著降低(P<0.05)。结论:脂联素、TNF-α和IL-6参与IR发生,健脾化痰汤可能通过调节IR脂肪细胞炎症细胞因子分泌,改善IR。Objective：To study the effect of serum containing Jianpi Huatan Decoction on the function of secreting adiponec- tin,tumor necrosis factor- α（ TNF-α） and interleukin- 6 （IL- 6） in insulin- resistant 3T3 -L1 adipocytes and explore its mechanism of improving insulin resistance. Methods：The 3T3 - L1 preadipocytes were cultured and differentiated to mature adipo- cytes in vitro. The cell models of insulin resistance were induced by dexamethasone. The normal group was established by adding 10% blank serum to the culture medium for normal adipocytes and the IR group by adding 10% blank serum to the culture medi- um for insulin - resistant adipocytes and the Chinese medicine group by adding 10% Jianpi Huatan Decoction serum to the cul- ture medium for insulin- resistant adipocytes and the western medicine group by adding 10% pioglitazone serum to the culture medium for insulin - resistant adipocytes. After cultured for 48 h, the glucose concentrations in the supernatants of the culture flu- id were measured with glucose oxidase method. The contents of adiponectin, TNF -α and IL - 6 in the supernatants of the culture fluid were measured with enzyme - linked immunosorbent assay （ELISA） method. Results ： Compared with the normal group, the amount of glucose consumption and the content of adiponectin were significantly decreased （ P 〈 0.05 ） and the contents of TNF -α and IL - 6 were markedly increased （P 〈 0.05 ） in the IR group. Compared with the IR group, the amount of glucose consump- tion and the content of adiponectin were remarkably increased （ P 〈 0.05 ） and the contents of TNF -α and IL - 6 were obviously decreased （P 〈 0.05） in the Chinese medicine group and the western medicine group. Conclusion： Adiponectin, TNF -α and IL - 6 could participate in the onset of insulin resistance. Jianpi Huatan Decoction could enhance adiponectin secretion of insulin - resistant adipocytes and degrade TNF -α and IL- 6 secretion to improve the insulin resistance.
