九味肝泰胶囊对急性酒精性肝损伤小鼠的防护作用及其机制

摘　　要：目的:研究九味肝泰胶囊对急性酒精性肝损伤小鼠的保护作用及其机制。方法:将雄性昆明种小鼠72只,随机分为正常对照组、模型组、硫普罗宁阳性对照组,九味肝泰胶囊低、中、高剂量组,连续灌胃给药8周。于第4周和第8周末分别单次灌胃白酒(56%V/V)10和16 ml·kg-1。观察九味肝泰胶囊对小鼠体质量和肝指数的影响,HE染色法观察肝组织的病理变化。测定各组小鼠血清中ALT、AST、ALP、GGT、TG、TC、LDL-C、HDL-C的水平,检测肝组织匀浆中SOD、MDA、GSHPx、GST水平和TNF-α、IL-1β含量的改变,并且RT-PCR法测定肝脏组织中TNF-α和IL-1β的mRNA表达。结果:九味肝泰胶囊中、高剂量组,能够显著降低小鼠肝指数,降低肝脏脂肪变性情况,小鼠血清中的ALT、AST、ALP和GGT都有明显回落,HDL-C含量升高,TC、TG和LDL-C的含量降低,肝组织中的SOD、GSH-PX和GST活性显著提高,MDA含量下降明显。此外,九味肝泰胶囊还抑制了小鼠肝组织中TNF-α,IL-1βmRNA的表达,降低了肝组织匀浆中TNF-α和IL-1β的含量。结论:九味肝泰胶囊对小鼠酒精性肝损伤有防治作用,可能与其调节脂质代谢,抗脂质过氧化,减少肝组织促炎因子的表达有关。OBJECTIVE To observe protective effects and mechanism of JiuWeiGanTai capsules （JWGT） against alcohol-in duced liver injuries in mice. METHODS Mice were randomly divided into six groups, namely, normal group, model group, tiopronin group, JWGT low dose group, medium dose group and high dose group. Changes of body weight and liver index were observed. Liver histopathology was revealed by HE staining. Serum and liver biochemical markers were measured. RESULTS Compared with model group, medium and high dose of JWGT reduced liver index in mice significantly, improved liver steatosis, decreased levels of ALT, AST, ALP, GGT, TC, TG and LDL-C in serum, increased level of HDL-C. Content of MDA in liver homogenate decreased significantly. Furthermore, the two dose groups increased activities of SOD, GSH-Px and GSHST obviously. Besides, JWGT could reduce TNF-α and IL-1β contents in liver tissue homogenate and inhibited expression of TNF-α and IL-1β mRNA in liver tissues. CONCLUSION This study provides a theoretical support that JWGT can protect acute liver injuries induced by alcohol.
