摘  要:目的:探讨他汀类药物对动脉中膜钙化的抑制作用及其对骨保护素(OPG)/NF-κB配体的受体(RANKL)的影响。方法:构建大鼠的主动脉中膜平滑肌细胞(SMC)的钙化模型,随机分为钙化组、他汀组和对照组。钙化组使用β-磷酸甘油、维生素C对SMC来进行钙化诱导;他汀组在上述基础上加入阿托伐他汀。用Von Kossa染色,钙离子含量测定,碱性磷酸酶(ALP)活性检测,骨钙素的Western Blot检测来判定细胞钙化的程度,用实时定量PCR检测细胞钙化过程中伴随的OPG和RANKL的表达情况。结果:加入他汀类药物后使得细胞钙化的程度得到减轻,同时细胞表达的OPG/RANKL比值也得到明显上升(均P<0.05)。结论:他汀类药物具有抑制动脉钙化的作用,其机制可能与提高OPG/RANKL的比值有关。Objective:To study the function of statins on the arterial calcification and its effect on the Osteopro- tegerin / Receptor Activator of NF- kB Ligand(OPG/RANKL). Method: To constitute the rat calcified smooth muscle cells model, the cells were divided randomly into the calcified group, statins group, and control group. For the calcified group, β-glycerophosphate and Vitamin C were added, for statins group, β-glycerophosphate, Vita- min C and atorvastatin were added; Von Kossa staining, Ca2+ content assay, ALP activity assay and osteocalcin assayed with Western Blot were used to check the level of cells calcification; Real Time PCR was used to check the expression of OPG and RANKL. Result: After stastins was added, the level of cells calcification decreased and the ratio of OPG / RANKL increased obviously (all P〈0.05). Conclusion: Statins could inhibit the arterial calcifica- tion, the mechanism was probably connected with its function of increase ration of OPG / RANKL.
