摘  要:从PDA平板上分离出一株能产生高活性淀粉酶和蛋白酶、且具广谱抗菌活性的芽孢杆菌AF1。结合单因素、正交实验和均匀实验,考察了不同碳源、氮源、培养温度等因素对AF1液体摇瓶培养浓度的影响,筛选出AF1的最佳液体生长培养基为:马铃薯20.0 g,蔗糖3.0g,蛋白胨2.5 g,尿素0.5 g,NaCl 1.0 g,蒸馏水100 m L;最佳培养条件为:温度为37℃,初始p H值为7.0,摇床转速为200 r/min。通过研究AF1对pH值、猪胆盐、人工肠液以及人工胃液的耐受性,发现其在pH值为3.0~9.0范围内均可生长,并且对猪胆盐、人工胃液和人工肠液表现出很强的耐受能力,表明AF1芽孢杆菌具有作为动物益生菌的开发潜质。A Bacillus species AF1 was isolated from the PDA medium. It was found that high-activity amylase and protease were produced by AFlwhen broad-spectrum antibacterial activities from AFI. Based on PDA medium, the effects of carbon sources, nitrogen sources, culture temperature and pH etc. on cell concentration were studied by combining single-factor, orthogonal test and uniform test. It was resulted that the growth medium was supplied with potato 20.0 g, cane sugar 3.0 g, peptone 2.5 g, urea 0. 5 g, NaC1 1.0 g, water 100mL and the optimum culture conditions included: temperature 37 ℃ , pH 7.0, rotation speed 200 r/min. Subsequently, the tolerance of AF1 to pH, cholate, simulated intestinal fluid and simulated gastric fluid were studied. It was showed that AF1 could grow well at pH3.0 -9.0 and it has good tolerance to cholate, simulated intestinal fluid and simulated gastric fluid. So it is indicated this Bacillus species AF1 could be used as a potential probiotic to serve livestock and poultry.
