附子中的新乌头碱对K562细胞增殖和凋亡的影响

摘　　要：目的研究附子中的成分新乌头碱作用于白血病K562细胞株对其生物学特性的影响。方法采用高效液相色谱法（HPLC）检测附子中是否含有新乌头碱和苯甲酰新乌头原碱。用新乌头碱标准品作用白血病K562细胞，电子显微镜下观察白血病细胞形态变化。流式细胞仪检测白血病细胞的细胞周期状态、凋亡的情况。结果高效液相色谱法检测出附子中含有新乌头碱。25—50mg／L的新乌头碱对K562细胞增殖有明显的抑制作用，并呈剂量和时间依赖性（P〈0．05）。K562细胞在形态学上有明显的凋亡变化。流式细胞仪检测白血病细胞的细胞周期和凋亡状态。S期比例上升，G。期比例降低（P〈0．05）。25～50mg／L的新乌头碱能不同程度地诱导K562细胞发生凋亡（P〈0．05）。结论附子中的新乌头碱具有抑制白血病K562细胞增殖、诱导其细胞凋亡的作用。Objective To study the biological effect of mesaconitine extracted from Fuzi （Radix Aconiti Lateralis Preparata Aconite） on human erythroleukemia （K.562） cells lines. Methods The contents of mesaconitine and benzoylmesaconine were determined by using HPLC. After K562 cells were treated with mesaconitine standard substance, the leukemia cell morphology was observed under an electron micro- scope microscope. And the cell cycle, apoptosis of leukemic cells were measured by flow cytometry. Re- suits Mesaconitine of 25 -50 mg/L significantly inhibited K562 cells proliferation in dose - and time - dependent manner（P 〈 0.05）. Apoptosis was induced by mesaconitine at the dose of 25 - 50 mg/L（P 〈 O. 05）. The ratio of cells at S phase increased and that of cells at G1 phase decreased（P 〈0.05）. Con- clusion Mesaconitine extracted from Fuzi inhibited proliferation and induced apoptosis of K562 cell line.
