摘 要:利用浸种法对转基因小麦后代进行卡那霉素筛选,种子在吸涨阶段所吸收的卡那霉素量决定着筛选效果,而种子的质量直接影响着卡那霉素的吸收量。针对不同遗传背景的小麦,为了省去每次筛选前对卡那霉素用量的调整,作者以与可变因素直接关联的种子质量为重点进行研究,探讨了一种简单、有效的筛选方法,并在小麦转基因后代检测中进行了验证。结果表明:以每克种子用150 mg/L卡那霉素溶液6 m L进行浸种,能够有效地对不同遗传背景的小麦进行鉴定。Kanamycin screening of transgenic wheat progeny was conducted by the method of seed-immersion, and the amount of kanamycin absorbed during imbibition stage of seeds determined the screening results, while the seed quality directly affected the kanamycin absorption. In order to omit the adjustment of kanamycin dosage before screening, in view of the different wheat genetic backgrounds, we focused on the seed quality that directly associated with variable factors, and seeked a simple and effective kanamycin screening method, at the same time, the method was verfied through dectection of transgenic wheat progeny. The results showed that different genetic backgrounds of wheat were effectively screening according per gram of seed immersed in 6 mL solution with 150 mg/L kanamycin.
