摘 要:目的观察β3整合素在脓毒症大鼠心肌组织中的表达,探讨其在脓毒症心肌损伤中的作用。方法采用腹腔注射大肠杆菌内毒素(LPS)制作脓毒症模型,在相对应的时间点采用左心室插管术测量心功能参数,留取心脏组织分别用免疫印迹(Western Blot)、免疫组化(IHC)检测β3整合素的表达以及HE染色、天狼猩红染色观察心肌组织形态学变化。结果心功能参数的改变主要发生在脓毒症早期,即左心室舒张压、左心室发展压、左心室压力上升的最大变化速率、左心室压力下降的最大变化速率、心肌耗氧指数与对照组相比较,差异有统计学意义(P<0.05),组织形态学观察实验组大鼠心肌组织细胞空泡变性,排列松散,结构紊乱,间质胶原纤维数量减少。Western Blot和IHC结果显示实验组β3整合素与对照组比较降低,且6h后较显著,差异有统计学意义(P<0.01)。结论在脓毒症心肌损伤的过程中,β3整合素表达改变显著,其可能参与了脓毒症心肌损伤的机制。Objective To investigate the dynamic changes of changes of expression changes of expressionβ3 integrin in septic rat hearts and its roles in sepsis induced myocardial injury. Methods Sprague-Dawle rats in experimental group were injected Escherichia coli endotoxin (LPS). At the corresponding time of points, the hearts were isolated for morphologic pathology and sirius red staining,and cardiac functions, including left ventricular developed pressure (LVDP), left ventricular pressure maximal rate of rise ( + dP/dt- max), left ventrieular pressure the maximum rate of descent ( - dP/dtmax), myocardial oxygen consumption index (SPIT* HR) and left ventricular end diastolic pressure (LVEDP) were measured using left ventrieular catheterization. The expression of β3 integrin in myoeardium were measured using western blotting and immunohistochemistry. Results Hemodynamic changes occurred in the early stage of sepsis. LVDP, + dP/dtmax, - dP/dtmax, SPTI * HR in experimental group decreased significantly, compared with those in control group (P 〈 0.05). Histopathology of the hearts in LPS group showed vacuolar degeneration, arrangement in loose, struc- tttral disorder, interstitial collagen fibers decreased. Western Blot and IHC showed that β3 integrin expression decreased in experimen- tal group, especially 6h after LPS injection( P 〈 0.01). Conclusions There were significant structural and functional changes in sep- sis-induced myocardial injury and β3 integrin involved in the mechanisms of myocardial injury in sepsis rats.
