摘 要:目的探讨针药结合疗法对帕金森病(PD)模型小鼠纹状体区多巴胺D1、D2受体活性的影响。方法将C57B L/6雄性小鼠随机分为正常组、模型组、针刺组、中药组、西药组及针药结合治疗组,每组再随机分为7天组(n=7)和14天组(n=7),应用腹腔内注射(MPTP)的方法制备PD模型小鼠。针刺组针刺百会、运动前区(双侧)、哑门;西药治疗组灌胃美多巴;中药组灌胃中药PD方;针药结合组针刺和中药同时施行,方法同中药组和针刺组。用免疫组化法观察治疗后PD模型小鼠脑内纹状体区多巴胺(DA)D1、D2受体活性的影响。结果针药结合组、西药治疗组小鼠脑内纹状体区DA的D1、D2受体活性与模型组比较,差异有统计学意义(P<0.01)。西药治疗PD模型小鼠,其模型小鼠纹状体区D1、D2受体活性改善最佳,最接近正常组D1、D2受体活性指标。针药结合治疗改善PD模型小鼠纹状体区D1、D2受体活性优于单纯中药治疗及针刺治疗。结论西药美多巴治疗PD,改善该病多巴胺D1、D2受体活性且具有明显优势,针药结合对PD的治疗优于传统单纯中药治疗及针刺治疗。Objective To investigate the effect of acupuncture combined with herbal medicine on striatal neurotransmitters dopamine (DA) of D1 and D2 receptor activity in mouse with Parkinson's disease(PD). Methods Twenty - eight adult male C57B L/6J mice were randomly divided into normal control group, PD model group, acupuncture group, herbal medicine group,western drug group, and acupuncture and herb combination group. Each group was divided into 2 subgroups,the 7 - day and 14 - day( n = 7). The PD models were established with intraperitoneal injections of MPTP in C57/BL mice. The mice in acupuncture group were applied to Baihui,section of premotor area(both sides),Yamen The mice in western drug group were treated with madopa. The mice in herbal medicine group were treated with PD prescription. The mice in acupuncture and herb combination group were treat with both acu- puncture and herbal medicine. The effect of striatal neurotransmitters DA of D1 and D2 receptor activity were measured by immuno- histochemical method after treatment. Rosults There were significant differences on striatal neurotransmitters DA of D1 and D2 re- ceptor activity among the acupuncture combined with herbal medicine group,the western drug group and the normal control group( P〈0. 01). The striatal neurotransmitters DA of D1 and D2 receptor activity demonstrated a significant improvement in western drug group com- paring to other groups. The striatal neurotransmitters DA of D1 and D2 receptor activity demonstrated a significant improvement in the acupuncture combined with herbal medicine group,which comparing to the acupuncture group and the herbal medicine group. Conci- usion Madopa can effectively improve the striatal neurotransmitters DA of D1 and D2 receptor activity. The acupuncture and herb combination group was more significant than the acupuncture group and the herbal medicine group in improvement of the striatal neu- rotransmitters DA of D1 and D2 receptor activity.
