USP22基因在人早孕期母-胎界面的表达及意义

摘　　要：目的:检测早孕期绒毛及蜕膜组织中泛素特异性水解酶22(USP22)基因和蛋白水平表达,以探讨USP22在母-胎界面的生理性调节作用。方法:分别采用免疫组织化学方法、荧光实时定量PCR法、Western blot检测20例早孕期正常妊娠妇女及15例不明原因复发性流产患者绒毛、蜕膜组织中USP22的定位、mRNA及蛋白表达水平。结果:USP22在早孕期绒毛和蜕膜组织中均呈阳性表达。在蜕膜基质细胞,USP22主要定位于胞质,少部分定位于胞核;在绒毛滋养细胞,USP22主要表达于绒毛细胞滋养细胞的胞质和胞膜。实时荧光定量PCR及Western blot检测显示绒毛及蜕膜组织中均可见USP22mRNA及蛋白的表达;与正常妊娠组比较,复发性流产组绒毛USP22mRNA及蛋白表达水平显著降低(P<0.05),蜕膜组织中USP22mRNA及蛋白表达水平差异无统计学意义(P>0.05)。结论:USP22在人早孕期母-胎界面表达,并参与了正常早孕期绒毛滋养细胞侵袭的调控和正常妊娠的维持。母-胎界面USP22表达下降可能是导致自然流产发生的原因之一。Objective： To investigate the localization and expression of USP22 in villus and decidua in the first trimester pregnancy. Methods. The localization, mRNA and protein expression levels of USP22 in villus and decidua from 20 cases of normal early pregnant women and 15 patients with unexplained recurrent spontaneous abortion were detected by immunohistochemistry, fluores- cence real-time quantitative PCR and Western blot. Results. Immunohistochemical showed that there were both expression in trophoblasts and decidual stromal cells of normal early pregnancy. USP22 mainly located in cytoplasm in decidual stromal cells and also mainly located in cytoplasm and cytomembrane of trophoblasts cells in villus. Real-time quantitative PCR and Western blot detection showed that the expression of USP22 mRNA and protein were in villi and decidua; Compared with normal pregnancy, USP22 mRNA and protein expression levels of villi in patients with recurrent spontaneous abortion decreased significantly（P〈0.05）, and there were no statis-tically significant difference in decidua （P〉0.05）. Conclusion： USP22 is expressed in the mater- nal-fetal interface, and participates in the regulation of trophoblast invasion in the normal early pregnancy and maintenance of normal pregnancy. The downregulation of USP22 expression in the maternal-fetal interface may be one of the causes of spontaneous abortion.
