摘  要:目的研究结缔组织生长因子(CTGF)在糖尿病大鼠肾组织中的表达及贝那普利的干预作用。方法应用链脲佐菌素诱导Wister大鼠造模,将大鼠随机分为3组:正常对照(N)组、糖尿病肾病(DN)组、贝那普利治疗(DNB)组。12周末时,常规方法检测各组大鼠血糖、尿总蛋白排泄量(UAlb)、血肌酐(Scr)、血尿素氮(BUN)、24 h尿白蛋白排泄率(UAER)及肾重/体重等指标,采用免疫组织化学技术检测大鼠肾组织中CTGF的表达水平。结果 12周末时,DN组UAlb、UAER较N组明显增加(P<0.01),Scr及BUN较N组明显增高(P<0.01);DNB组UAlb、UAER、Scr及BUN较DN组显著减少,DNB组肾皮质CTGF的表达较DN组降低(P<0.01或P<0.05)。结论贝那普利可能通过抑制高血糖、糖基化终末产物(AGEs)抑制CTGF的表达,特异性阻断CTGF将延缓DN的发展。[ Objective ] To investigate the effects of Benazepril on expression of connective tissue growth factor (CTGF) in renal tissue of diabetic rats. [ Methods ] The indexes and CTGF in renal tissue from three groups of rats, which were healthy rats (N group), diabetic nephropathic rats induced by streptozotocin (STZ) (DN group), and dia- betic nephropathic rats treated with Benazepril (DNB group), were measured at the end of 12 weeks after induced. The indexes, including the mass index of body weight, blood glucose, urinary albumin (UAlb), blood urea nitrogen (BUN), 24h urinary albumin excretion rate (UAER), serum cretinine (Scr) and ratio of kidney weight and body weight, were measured by conventional methods. CTGF in renal tissue was measured by immunohistochemical tech- nique. [Results] Blood glucose, UAlb, UAER, BUN, Scr, renal expression of CTGF were significantly increased in DN group compared to those in N group (P 〈 0.01). UAlb, UAER, BUN and renal expression of CTGF were signifi- cantly decreased in DNB group compared to those in DN (P 〈 0.01, P 〈 0.05). [Conclusions] These findings sup- port our hypothesis that Benazepril may inhibit the expression of CTGF by controlling high glucose and AGEs. We contend that blocking CTGF will delay the progression of Diabetic Nephropathy.
