摘 要:目的观察吸入麻醉药异氟烷对大鼠条件性味觉厌恶(CTA)记忆的获取、巩固和再巩固3个不同阶段的影响。方法将72只健康成年雄性Sprague-Dawley大鼠随机分入记忆获取组、记忆巩固组和记忆再巩固组,每组24只;每组均再分为异氟烷暴露组和空氧混合气体暴露组2个亚组,每亚组12只。所有大鼠均予CTA训练使其对糖精钠溶液产生厌恶记忆。3组大鼠分别在CTA记忆的获取、巩固和再巩固阶段予1个最低肺泡有效浓度(MAC)异氟烷(体积分数为0.015)或空氧混合气体(空气和氧气的体积分数分别为0.7和0.3)暴露2h。CTA训练后24、48和72h连续3次检测大鼠的糖精钠溶液厌恶指数(AI)以观察异氟烷暴露对大鼠CTA记忆不同阶段的影响。结果记忆获取组中,异氟烷暴露亚组在CTA测试期连续3d的糖精钠溶液AI均显著低于空氧混合气体暴露亚组(P值分别<0.05、0.01),且随时间延长差异更为明显。记忆巩固组两个亚组间CTA测试第1天的糖精钠溶液AI的差异无统计学意义(P>0.05),第2、3天异氟烷暴露亚组显著高于空氧混合气体暴露亚组(P值均<0.05)。记忆再巩固组中,两个亚组间在CTA测试期连续3d的糖精钠溶液AI的差异均无统计学意义(P值均>0.05)。结论异氟烷可损害大鼠CTA记忆的获取,并增强其巩固过程,但对其再巩固无显著影响。Objective To investigate the influence of isoflurane on acquisition, consolidation and reconsolidation of conditioned taste aversion (CTA) memory in rats. Methods Seventy-two male adult Sprague- Dawley rats were randomly divided into 3 groups (n = 24): acquisition group, consolidation group and reconsolidation group. Then the rats in each group were further divided into 2 subgroups (n = 12): isoflurane subgroup and air+ O2 subgroup. An aversion to saccharin sodium water was developed through CTA training. In the process of acquisition, consolidation and reconsolidation of CTA training, rats of each group were exposed to either isoflurane ( 1 minimum alveolar concentration [EMAC], volume fraction was 0. 015) or air+ O2 (volume fraction of air and O2 were 0. 7 and 0. 3, respectively) for 2 h. Aversion indexes (AI) of saccharin sodium water were consecutively measured 24 h, 48 h and 72 h after CTA training. Results In acquisition group, the rats in isoflurane subgroup showed lower AI than air+ O2 subgroup in consecutive 72 h after CTA training (P〈0.05 or P〈0.01), with a remarkable difference as time going. In consolidation group, there was no significant difference in the AI on day 1 of test between two subgroups (P〉0.05) ; however, At of isoflurane subgroup were significantly higher than those of air+ Oz subgroup in the next 2 days of the test (both P〈0.05). In reconsolidation group, there were no significant differences in the AI in the consecutive 3-day test between two subgroups (all P〉0.05). Conclusion Isoflurane can impair the acquisition of CTA and enhance the consolidation of CTA, but it will not affect the reconsolidation of CTA.
