摘  要:目的研究氧化应激在背根神经节慢性压迫(CCD)模型致大鼠镜像痛中的作用。方法选取雌性Sprague-Dawley(SD)大鼠建立L4、L5两节段CCD模型(CCD模型组),并设立假手术对照组(10只),应用von Frey纤维丝测定双侧后足机械痛缩足阈值(PWMT)。术后第7天,筛选出CCD模型组发生镜像痛大鼠30只,按照5mL/kg标准分别予腹腔内注射氧自由基清除剂N-叔丁基-α-苯基硝酮(PBN)100mg/kg(CCD+PBN100亚组,10只)、PBN 30mg/kg(CCD+PBN30亚组,10只)和0.9%氯化钠溶液(CCD+NS亚组,10只)。记录并比较各组大鼠双侧后足在给药前0.5h和给药后0.5、1、2、4、6、8、24h时的PWMT值和镇静评分。结果CCD模型组术后第1、3、5、7天压迫侧和损伤对侧的PWMT值均显著低于同组同侧术前(P值均<0.05),假手术组术后第1天压迫侧和损伤对侧的PWMT值均显著低于同组同侧术前(P值均<0.05)。CCD模型组术后第1、3、5、7天压迫侧和术后第3、5、7天损伤对侧的PWMT值均显著低于假手术组同侧同时间(P值均<0.05)。CCD模型组术后1、3、5、7天损伤对侧的PWMT值均显著高于同组压迫侧同时间(P值均<0.05)。CCD+PBN100亚组注射后0.5、1、2h压迫侧和损伤对侧的PWMT值均显著高于同亚组同侧注射前和CCD+PBN30亚组同侧同时间(P值均<0.05),CCD+PBN30亚组注射后0.5、1、2h压迫侧和注射后0.5、1h损伤对侧的PWMT值均显著高于同亚组同侧注射前和CCD+NS亚组同侧同时间(P值均<0.05)。CCD模型组3个亚组的镇静评分均为0分,3组间差异均无统计学意义(P值均>0.05)。结论腹腔注射氧自由基清除剂PBN可以缓解大鼠CCD引发的镜像痛行为。Objective To investigate the possible role of oxidative stress in mirror-image pain (MIP) induced by chronic compression of dorsal root ganglia (COD). Methods Neuropathic pain was induced by COD operation in female Sprague-Dawley rats. Meanwhile, 10 rats received Sham operation. Paw withdrawal mean threshold (PWMT) was measured by von Frey filament. Thirty COD rats in which MIP had been developed were equally randomized into three groups 7 d after COD operation. PWMT and sedation scores were assessed before and after phenyl N-tert-butyl nitrone (PBN, an oxygen free radical scavenger, 100 mg/kg, 30 mg/kg) or normal saline was injected intraperitoneally with 5 mL/kg solution. Results Both ipsilateral and contralateral PWMT on day 1, 3, 5, and 7 after CCD operation were significantly lower than those before the operation (all P〈0.05). Ipsilateral and contralateral PWMT on day 1 after sham operation were significantly lower than those before the operation (both P〈0.05). Ipsilateral PWMT on day 1, 3, 5 and 7 and contralateral PWMT on day 3, 5 and 7 after CCD operation were significantly lower than those after sham operation (all P^0.05). Contralateral PWMT on day 1, 3, 5 and 7 after COD operation were significantly higher than ipsilateral one (all P^0.05). Ipsilateral and contralateral PWMT at 0.5 h, 1 h, 2 h after PBN 100 mg/kg injection were significantly higher than those before injection ( all P〈0.05). Ipsilateral PWMT at 0.5 h, 1 h, 2 h after PBN 30 mg/kg administration and contralateral PWMT at 0.5 h and 1 h after PBN 30 mg/kg administration were significantly higher than those before administration (all P〈0.05). Ipsilateral and contralateral PWMT at 0.5 h, 1 h and 2 h after PBN 100 mg/kg injection were significantly higher than those after PBN 30 mg/kg injection (all P^0.05).
