摘  要:目的观察使用新型亚甲蓝病毒灭活套材对血浆组分质量指标的影响。方法随机领取100m L/U的新鲜冰冻血浆50U,连接至亚甲蓝病毒灭活套材,溶解内置的冻干亚甲蓝,置病毒灭活箱内照射,照射结束后,通过亚甲蓝白细胞滤器滤除亚甲蓝。分别于照射前、照射后及过滤后留取样品,置-20℃冰箱保存。检测各样品FⅤ、FⅧ、FⅨ、FⅪ、FⅫ、FIB-C、AT、VWF-Ag、VWF-Rco、F1+2、C3a、C5a、IL-6及IL-8的含量。结果经病毒灭活处理后血浆凝血因子Ⅴ∶C、Ⅷ∶C、Ⅸ∶C、Ⅺ∶C、Ⅻ∶C、FIB-C、VWF-Rco的回收率分别为81.70%、71.55%、81.69%、87.78%、90.93%、71.82%和85.05,AT和VWF-Ag的含量未受影响。F1+2、C3a、C5a、Il-6和IL-8分别下降了9.03%、3.25%、19.01%、20.37%、14.55%。结论使用新型亚甲蓝病毒灭活套材处理后,血浆凝血因子及其他血浆组分影响在可接受范围内,符合国家全血及成分血质量要求以及临床安全输血的需求。Objective To observe the influence of virus inactivation of fresh-frozen plasma in the new methylene blue system on the quality of plasma components. Methods A total of 50 fresh frozen plasma units ( 10OraL each) were connect- ed to the new methylene blue system. The plasma passed through tubing containing a MB dry pill that released as the plasma flowed through tile tubing into the illumination bag. The illumination bag was subjected to photochemical virus inactivation. Then, the plasma was passed through the filter that removed the residual methylene blue and white blood cells. Plasma sampies that were remained in the various stage were immediately frozen in -20℃ refrigerator. All samples were analyzed for the following contents: Ⅴ : C, Ⅷ: C, Ⅸ: C, Ⅺ: C. Ⅻ: C. FIB-C, AT, VWF-Ag, VWF-Rco, F1 + 2, C3 a, C5a 、 IL-6 and IL-8. Results After methylene blue photochemical treatment, the recovery rate of coagulation factors were 81.70% F V, 71.55% Fvm, 81.69% FⅨ, 87.78% FⅪ, 90. 93% FⅫ, 71.82% FIB-C, 85.05% VWF-Rco. No losses were seen for VWF-Ag and AT. A decrease was seen in other components: 9.03% F1 +2, 3.25% C3a, 19. 01% C5a, 20. 37% IL-6 and 14. 55% IL-8. Conclusion The effect of photochemical treatment using the new methylene blue system on coagulation factors and other components in plasma is in an accepted range, and meets the requirements of whole blood and blood components quality and the clinical demand for safe blood transfusion.
