摘  要:为了研究利巴韦林对感染猫细小病毒(FPV)细胞的影响,试验将猫肾细胞(F81细胞)随机分为对照组、病毒组、药物组,采用MTT法检测细胞的生长曲线,Hochest 33258染色观察细胞的细胞核形态,流式细胞仪检测细胞周期。结果表明:Hochest 33258染色试验中药物组与病毒组差异不显著(P>0.05);药物组细胞的增殖活性始终高于病毒组,但低于对照组;流式细胞仪检测利巴韦林治疗后48小时G0/G1期细胞增多,S期细胞减少,G2/M期细胞变化不大;72小时G2/M期细胞增多;药物组与病毒组细胞的流式检测在G1峰的左侧均出现典型的亚G1峰,但各时间点药物组细胞的凋亡指数均明显低于病毒组。说明利巴韦林能抑制FPV诱导的细胞凋亡,具有一定的治疗指导意义。To study the effect of ribavirin on the feline parvovirus (FPV) cells, the FS1 cells were used and divided into a control group, virus group, and drug group. The cell growth curves were detected by MTT assay, and the morphology of cell nuclei was observed by Hochest33258 staining, and the cell cycles were detected by flow cytometer. The results of the MTT assay showed that the proliferation activity of the cells in the drug group was consistently higher than that of the virus group, but lower than that of the control group. The results of morphological obser- vation showed that there was no significant difference ( P 〉 0.05 ) between the drug group and virus group in the Hochest33258 staining test. The testing results of flow cytometer showed that the GO/G1 phase cells increased, and S - phase cells decreased, and G2/M phase cells changed little after 48 h of ribavirin treatment in the drug group ; the G2/M phase cells increase after 72 h of ribavirin treatment. The cells in the drug group and virus group presented a typical sub - G1 peak on the left of GI peak, but the apoptosis index of the drug group was significantly lower ( P 〈 0. 05 ) than that in the virus group at each time point. The results indicate that ribavirin can inhibit the apoptosis induced by FPV, and it has a certain significance for guiding the treatment of FPV.
