煎煮时间对钩藤治疗肾性高血压有效成分钩藤碱含量析出的影响

摘　　要：目的:分析不同煎煮时间钩藤中钩藤碱析出含量,以及不同煎煮时间钩藤对治疗大鼠肾性高血压的影响。方法:按不同时间水煎煮钩藤(5,10,15,30min),测定钩藤碱的含量。60只SD大鼠采用结扎左肾血管法进行肾性高血压大鼠模型造模。将造模成功的大鼠随机分为模型组、不同煎煮时间(5,10,15,30min)钩藤提取液组,每组各10只大鼠。另设空白组大鼠10只。灌胃给药1次/d,2ml/次,连续给药6周。末次给药2h后,使用无创血压测量系统对各组大鼠血压进行测量。结果:钩藤碱煎煮15min的钩藤碱含量最高。与空白组相比,模型组的血压明显升高(P<0.05)。与模型组比,钩藤碱给药组的大鼠的血压较模型组的低(P<0.05),其中煎煮15min组血压降低较为明显。结论:钩藤中钩藤碱具有较好的降血压效果,但钩藤随着煎煮时间的延长,生物碱煎出量有所下降,影响临床疗效。Objective： To study the content of rhy under different different decoction time of uncaria tomentosa on the treatment of ods. Uncaria tomentosa had been decocted for 5 minutes, 10 m respectively, then the content of rhy was determined respective in ly decoction time, and the effect of renal hypertension in rats. Meth- utes, 15 minutes, and 30 minutes Models of 60 SD rats with renal hypertension were established by the ligation of the left renal artery method. The successfully-established model rats were randomized into the model group, groups of different decoction time of uncaria extract （for 5 min, 10 min, 15 min, and 30 min） with 10 rats each, and another 10 rats were selected as the blank group. Administered orally once a day, 2 ml a time, for six consecutive weeks. 2 h after the last administration, non-invasive blood pressure measuring system was used to measure the blood pressure in rats. Results： The content of rhy reached the highest when uncaria had been decocted for 15 min. Compared with the blank group, the blood pressure increased more significantly in the model group （P〈0.05）. Compared with the the model group, the blood pres-sure in rats of the rhy administrated group was lower （P〈0.05）, of which the blood pressure reduced more significantly when uncaria had been decoeted for 15 rain. Conclusion： Rhy in unearia tomentosa has good hypotensive effect, but the decoction time should not be for long. With the pro longing of the decocting time, the content of the rhy will decreased to some extent, and it will af fect the clinical outcome.
