摘  要:目的在组织和细胞水平探讨KDM1A的表达水平,并分析其对胃癌侵袭转移的影响。方法通过Western Blot技术检测胃癌组织及胃癌细胞中KDM1A蛋白的表达;通过Transwell侵袭实验检测胃癌细胞的侵袭性;利用RNA干扰技术抑制胃癌细胞BGC823中KDM1A的表达。结果在胃癌组织及胃癌细胞株中KDM1A蛋白质表达升高;胃癌细胞的侵袭转移能力与KDM1A蛋白表达量之间均存在正相关关系;siRNA-KDM1A可在体外干扰胃癌细胞BGC823中KDM1A蛋白表达,并影响BGC823的侵袭转移能力。结论 KDM1A在胃癌组织及细胞中表达上升,在体外实验中抑制KDM1A表达,可抑制胃癌细胞的侵袭转移。Objective To explore the expression level of KDM1A in tissues and cell lines, and to analyze its impact on the invasion and metastasis of gastric cancer. Methods Used western blot to detect KDM1A protein expression in gastric cancer tissue and gastric cancer cell lines. Used transwell invasion assey to detect gastric cancer cell lines in vitro. Used RNA interference technique to silence KDM1A in gastric cancer cell line BGC823. Results There was higher KDM1A protein expression in gastric cancer tissues and cell lines. There was positive correlation between protein KDM1A expression and invasive and metastatic ability of gastric cancer cell lines, siRNA-KDMIA interfered BGC823 KDM1A protein expression in gastric cancer cells in vitro, and affected BGC823 ability of invasion and metastasis. Conclusion There is higher KDM1A expression in gastric cancer tissues and cell lines, and the decrease of KDM1A expression in- hibits the invasion and metastasis in vitro.
