摘 要:目的观察不同来源干细胞移植治疗大鼠创伤性脊髓损伤后的免疫反应差异。方法SD大鼠68只随机分成四组:羊膜上皮细胞(HAECs)移植组(20只),脐带间充质干细胞(HUCMSCs)移植组(20只),骨髓间充质干细胞(BMSCs)移植组(20只),PBS对照组(8只)。测定血清中IL-2及IL-10,检测移植局部CD3+细胞浸润的情况。结果在移植后1周和1、2、3个月时,检测到移植组大鼠各个时间点静脉血中IL-2水平较对照组升高,而IL-10水平较对照组下降(P<0.05)。BMSCs组大鼠在移植后各时间点静脉血中IL-2水平较HUCMSCs组和HAECs组升高,IL-10水平下降(P<0.05)。四组在急性期(细胞移植后1周)移植局部均有大量CD3+细胞浸润;而在移植的慢性期(细胞移植后3个月)BMSCs组CD3+细胞浸润仍高于其他三组(P<0.05)。结论异体干细胞移植后可发生急、慢性免疫排斥反应。胚胎源性干细胞较大鼠BMSCs有更低的免疫源性。Objective To observe the differential immunological responses of rats after spinal cord injury to transplanated stem cells from different sources .Methods Sixty‐eight SD rats with spinal cord injury were randomly divided into 4 groups of A (transplanated with human amniotic epithelial cells ,20 rats ) ,B (transplanated with umbilical cord mesenchymal stem cells ,20 rats ) , C(transplanated with bone marrow mesenchymal stem cells ,20 rats) and D (PBS control ,8 rats) . Serum levels of IL‐2 and IL‐10 were detected and the CD3+ cell infiltration was measured in one week ,and 1 ,2 and 3 months after transplantation .Results Serum level of IL‐2 after transplantation was obviously higher ,but that of IL‐10 was lower ,in groups of A ,B and C than that in group D (P〈0 .05) ,which were more in group C than those in groups of A and B(P〈0 .05) .The quantity of CD3+ cells infiltration was obviously seen in all groups in one week after transplantation ,which in 3 months was still more in group C than that in the other three groups(P〈0 .05) .Conclusion Systemic and local immunological rejection in acute or chronic periods of stem cells transplantation can be observed in rats with spinal cord injury ,which are lower in rats transplanated with human amniotic epithelial cells than that with bone marrow mesenchymal stem cells .
