摘  要:采用HPLC法同时测定不同产地博落回各部位血根碱、白屈菜红碱含量.采用Welch Material XB-C18分析色语柱(4.6×250mm,5μm),以(A)体积分数为0.1%冰乙酸水溶液-(B)乙腈梯度洗脱(0~24min,B的体积分数为10%~46%)为流动相,流速为1mL/min,柱温25℃,检测波长270nm.结果在一定质量浓度范围内血根碱、白屈菜红碱含量测定方法的线性关系良好,相关系数分别为0.9997、0.9993;平均回收率分别为95.6%、95.9%,RSD分别为1.06%、0.6%(n=3).该方法操作简便、准确、灵敏度高,适合于博落回中血根碱、白屈菜红碱含量的测定,不同部位两种生物碱含量均存在明显差异,不同采集地点的样品之间两种生物碱含量也存在一定差异.To establish a HPLC method for the determination of sanguinnarine and chelerythrine contents in Macleaya cordata,so as to investigate their contents in different parts of plant individuals collected in different areas. The analysis was performed on a Welch Material XB-C18 column( 4. 6 × 250 mm,5 μm) with gradient( A) 0. 1% acetic acid-( B) acetonitrile( 0 ~ 24 min,10% ~ 46% B) as mobile phase. The flow rate was 1 m L / min. The column temperature was 25 ℃ and the detection wavelength was 270 nm. A good linearity was obtained and the correlation coefficient were 0. 999 7 and 0. 999 3 respectively. The average recoveries were 95. 6% and 95. 9% with corresponding RSDs of 1. 06%,0. 6%,respectively( n = 3). The method is simple and accurate with a good sensitivity. It can be used for determining sanguinnarine and chelerythrine in Macleaya cordata. The difference of contents of sanguinnarine and chelerythrine in different parts from individual plant material of Macleaya cordata is significant. The contents of sanguinnarine and chelerythrine of M. cordata from different areas are also different.
