摘 要:目的:探讨子痫前期(PE)大鼠胎盘组织白细胞介素-6(IL-6)表达异常对胎鼠肺发育的影响。方法:将16只孕鼠随机分为模型组(PE早产组)和对照组(非PE早产组)各8只,模型组使用内毒素建立PE动物模型,对照组给予等量生理盐水。两组孕鼠妊娠第20天时剖腹产取出胎鼠及胎盘,采用免疫组织化学SABC染色法和免疫印迹法检测胎盘组织IL-6表达水平。两组胎鼠饲养14 d后取肺组织制成切片,进行苏木精-伊红(HE)染色,放射状肺泡计数(RAC)分析肺泡发育情况,免疫印迹法检测肺组织血管内皮生长因子(VEGF)表达水平。结果:模型组孕鼠胎盘组织IL-6表达水平高于对照组,胎鼠RAC多于对照组,胎鼠肺组织VEGF表达水平高于对照组(P均<0.05)。结论:PE大鼠胎盘组织IL-6的表达增高,进而促进胎鼠肺组织VEGF的表达和肺泡的形成,但胎鼠肺泡缺乏表面活性蛋白而导致肺发育不成熟。Objective: To investigate whether the level change of interleukin-6 (IL-6) in preeclampsia placenta is sufficient to cause abnormalities of fetal rat lung development during infancy. Methods: Endotoxin was used to build maternal preeclampsia model for PE preterm group, non PE preterm group used saline. Two group preterm rats ( each group 8 rats) were all delivered by e-section at 20 days of gestation. Expression of IL-6 in plaeenta was tested through immunohistochemistry and western blot. Preterm rats were observed through 14 days of age for study. The pathological changes of the fetal rat lung were observed by HE staining and Radical Alveolar Counter. The protein expression level of vascular endothelial growth factor (VEGF) was detected by western blot . Results: Compared to non PE preterm group, the expressions of IL-6 in PE preterm group were widely higher in placenta. Increased alveolar count was shown in PE preterrn group, and the increased VEGF expression can also be detected in PE preterm group (all P〈0.05 ). Conclusion: Preeelampsia earl promote the IL-6 expression in placenta and accelerate the vaseularization of the fetal lung and inerease the alveolar count, but the alveoli are not mature with lacking of surfactant protein.
