电针对吗啡戒断大鼠NAc脑区Ca^(2+)浓度和CaM活性的影响

摘　　要：目的:研究电针对吗啡戒断大鼠空间学习记忆能力及对NAc脑区Ca2+浓度和CaM活性的影响。方法:采用逐日递增法,背部皮下注射吗啡5天后立即给予纳洛酮催促戒断,建立吗啡戒断大鼠模型,然后对戒断症状评分。采用Morris水迷宫测试大鼠空间学习记忆能力;采用流式细胞技术检测不同组别的细胞内Ca2+浓度、CaM的变化。结果:电针组大鼠逃避潜伏期较模型组明显缩短(P<0.01);电针组与模型组比较显著增加穿越平台次数(P<0.01)。结论:电针治疗可明显改善吗啡戒断后大鼠空间学习和记忆能力;电针组大鼠脑内Ca2+浓度、CaM较模型组明显升高。Objective：To study the effect of electroacupuncture on spatial memory and learning and detect Ca2＋concentration and activity of CaM of NAc area in morphine withdrawal rats.Methods：Make models with mor-phine withdrawal.Adult male rats were chronically injected with cumulative subcutaneous （s.c.） doses of mor-phine within five days,and the precipitated withdrawal symptoms were scored after naloxone injection.Morrirs water maze test was performed for the rats to test their spatial memory,and intracellular Ca2＋concentration and activity of CaM in different groups were detected by flow cytometry.Results：Compared with the model group , electroacupuncture group showed a significant decrease of time to reach platform and increase of the frequency to cross the platform.（ P〈0.01）.Ca2＋concentration and activity of CaM of NAc area in electroacupuncture group were significantly higher than those in the model group.（ P〈0.01） .Conclusion：These findings suggest that electroacupuncture significantly improve spatial learning and memory in morphine withdrawal rats, which may be associated with an increase in brain NAc Ca2＋concentration and activity of CaM after electroacupunc-ture.
