摘 要:目的对2014年安徽省一起人感染布鲁菌疑似患者进行表型和分子生物学检测,以明确诊断。方法对患者进行流行病学调查,采用血培养法对经试管凝集试验检测为布鲁菌抗体阳性的患者和病羊血液进行荧光PCR及细菌分离培养,应用传统方法和分子生物学方法对可疑菌落进行种型鉴定。结果 136份高危者及疑似布病患者血清抗体筛查22份阳性,31份患者、病羊和外环境采样标本经荧光PCR检测5份阳性。2株安徽省分离株经传统方法和种型特异性PCR均鉴定为羊种生物3型布鲁菌。结论从患者和病羊均分离出羊种生物3型布鲁菌,结合流行病学资料分析,安徽省此次疫情由人直接接触病羊而感染布鲁菌所致。Objective To detect the phenotype and molecular biological characteristics of a case of suspected patient with Brucella in Anhui in 2014,so as to identify the diagnosis. Methods The patient was performed on the etiological investigation,and blood culture method was used to isolate and culture Brucella srtains from the blood of anti- Brucella antibody positive goats and patients detected with standard agglutination test( SAT),and conventional and molecular techniques were used to identify the suspicious bacteria strains. Results In the antigen screening of 136 high risk patients and the suspected patients with brucellosis,22 were detected to be positive. A total of 31 samples from patients,sick sheep and external environmental sampling were detected by the real- time fluorescence PCR( RT- PCR). and 5 were detected to be positive. Besides,2 strains were identified as B. melitensis biotype 3 by the conventional test and species- specific PCR. Conclusion 3 bivar Brucella was isolated from both patient and sick goat. and combined with the analysi epidemiologic data,it could be concluded this epidemic was caused by the direct human contact with sheep with Brucella.
