摘 要:目的了解2014年鞍山地区麻疹暴发疫情的优势基因型别,并分析其与H1a亚型参考株Chin9322平均核苷酸和氨基酸的同源性。方法采集2014年鞍山市暴发的疑似麻疹患者的咽拭子标本,使用WHO推荐的vero-SLAM细胞进行麻疹病毒分离,用反转录-聚合酶链反应(RT-PCR)扩增麻疹病毒分离株的N基因羧基末端450 bp片段。对扩增产物的核苷酸序列与WHO各基因型参考株基于N基因构建的基因亲缘性关系树,分析麻疹病毒基因型。结果150份咽拭子标本中分离到26株麻疹病毒毒株,经对分离株的核蛋白(N)碳末端450个核苷酸进行序列测定和亲缘性关系比较,26株麻疹病毒均属于H1基因型的H1a亚型,其中核苷酸和氨基酸之间的同源性分别为98.2%~100.0%和98.0%~100.0%。结论 2014年鞍山地区麻疹暴发疫情是由麻疹病毒H1基因型的H1a亚型引起的流行,本次流行期内的麻疹病毒几乎是同源的,在病毒的传播过程中几乎不发生基因变异。Objective To investigate the predominant genotype of measles in the outbreak of Anshan area in 2014,and analyze the homology of it with the average nucleotide and amino acid of H1 a subtype reference strain Chin9322. Methods Collecting throat swab samples of suspected measles patients outbreaked in Anshan city in 2014. Isolating the measle virus through using the vero- SLAM cells recommended by WHO,and increasing the nucleoprotein( N) gene C- terminal 450 bp fragment of measles virus isolates by using reverse transcriptase- polymerase chain reaction( RT- PCR). Analyzing the measles virus genotype of nucleotide sequence of amplification products and gene phylogenetic tree of WHO genotype reference strains based on N gene.Results 26 strains of measles virus were isolated from 150 throat swab specimens. Through sequencing and comparing phylogenetic relationship of 450 nucleotides at the carbon treminal of nucleoprotein( N) of the isolates,26 strains of measles virus belonged to the H1 a subtype of H1 genotype. And the homology between the nucleotide and amino acid were respectively98. 2%- 100. 0% and 98. 0%- 100. 0%. Conclusion The measles outbreak in Anshan region in 2014 was caused by H1 a subtype of the measles virus H1 genotype. The measles viruses in the epidemic period are almost homologous,and there are scarcely any genovariations in the spreading process.
