摘  要:目的探讨姜黄素对大鼠心肌缺血再灌注损伤的保护作用。方法将60只成年健康Wistar大鼠采用随机数字表法分成6组,即假手术组、缺血30 min组、再灌注1、2 h组、姜黄素+再灌注1、2 h组,每组10只。应用放射免疫方法检测血清内皮素-1(ET-1)水平;反转录-聚合酶链反应及蛋白免疫印迹法检测大鼠心肌组织ET-1 mRNA及蛋白表达水平。结果与假手术组比较,缺血30 min组、再灌注1、2 h组、姜黄素+再灌注1、2 h组大鼠血清ET-1和心肌组织ET-1 mRNA及蛋白表达水平均显著升高(P<0.05);与缺血30 min组比较,再灌注1、2 h组、姜黄素+再灌注1、2 h组大鼠血清ET-1和心肌组织ET-1 mRNA及蛋白表达水平显著升高,且再灌注2 h组显著高于再灌注1 h组(P<0.05);姜黄素+再灌注1、2 h组较再灌注2 h组大鼠血清ET-1和心肌组织ET-1 mRNA及蛋白表达水平显著下调(P<0.05)。结论 ET-1在急性心肌缺血再灌注损伤缺血和再灌注阶段表达上调,姜黄素预处理可下调其表达。Objective To explore the protective effect of curcumin on acute myocardial ischemia and reperfusion injury rats. Methods Sixty Wistar rats were randomized divided into sham operation group,ischemic 30 minutes group, reperfusion 1 hour group, reperfusion 2 hours group, curcumin + reperfusion 1 hour group, and curcumin + reperfusion 2 hours group by ran- dom digits table,with 10 rats in each group. The level of endothelin-1 in blood serum was determinated by radioimmunoassay method ; the expression of ET-1 mRNA and protein in muscle tissue were detected by reverse transcription-polymerase chain re- action and Western blot method. Results The level of endothelin-1 in blood serum, the expression of ET-1 mRNA and protein in muscle tissue in sham operation group were significantly lower than other groups ( P 〈 0.05 ). Compared with ischemie 30 minutes group, the levels of endothelin-I ,the expression of ET-1 mRNA and protein in reperfusion one hour group, reperfusion two hours group, curcumin + reperfusion one hour group and curcumin + reperfusion two hours group increased significantly (P 〈 0.05 ) ;and the reperfusion two hours were significantly higher than reperfusion one hour( P 〈 0.05 ). The level of endo- thelin-1 ,the expression of ET-1 mRNA and protein in cureumin + reperfusion one hour group and Cureumin + reperfusion two hours group were significantly lower than reperfusion two hours group ( P 〈 0.05 ). Conclusion The endothelin-1 level in- crease in myocardial ischemia and reperfusion injury in rats and the curcumin can reduce its level.
