摘  要:目的:超声影像动态观察小鼠横向主动脉缩窄模型的心功能变化过程,探讨此模型法能否模拟临床早期心力衰竭病理过程。方法:100只,C57/B6L,雄性小鼠,体质量(22±3)g,随机分成假手术组(15只)和模型组(85只)。分别于手术后第1周分析结扎处血流速度峰值,第1,2,3,4,5,6及第8周使用小动物超声影像分析左心室心肌收缩功能和室腔变化。结果:术前小鼠主动脉弓内径为(1.161±0.025)mm,手术缩窄后主动脉弓横截面积为缩窄前横截面积的(19±1)%。模型第1周,超声检测缩窄处的血流峰值为(2279±54)mm/s,为假手术组血流峰值的2.85倍(P<0.01)。第4周,模型组左心室质量/体质量比值显著增加,超声影像学分析显示,左心室前壁舒张末厚度增加42.1%,左心室后壁舒张末厚度增加31.1%,左心室重量增加56.0%,与假手术组相比(分别P<0.01);组织切片WGA染色结果显示模型组心肌细胞横截面积明显增大,与假手术组相比(P<0.05);第8周,模型组小鼠左心室射血分数降低32.3%,短轴缩短率降低38.6%,与假手术组相比(分别P<0.01)。结论:横向主动脉缩窄第4周特征为左心室肥厚、心肌肥大,模拟了临床上心力衰竭早期的病理生理过程。Objective: Dynamic observation of ultrasound imaging to transverse aortic constriction model,which pathologic process simulated cardiac hypertrophy early in clinical heart failure. Methods: 100 male C57 / B6 L mice,weighting( 22 ± 3) g,were randomly divided into sham group( 15 mice) and surgical group( 85 mice). One week later,ultrasound image analysis blood flow peak velocity at ligation. Ultrasound imaging used to analysis left ventricular function and cavity changes at 1st,2nd,3th,4th,5th,6thand 8thweek respectively.Results: The inner diameter of the aortic arch was 1. 161 mm ± 0. 025 mm before ligation. The aortic arch cross-sectional area of model group is( 19 ± 1) % compared with sham group. One week after ligation,blood flow peak velocity at ligation can reach( 2279 ± 54) mm / s,which is 2. 85 fold of sham group( P〈0. 01). At four weeks after surgical,the left ventricular weight to body weight ratio of surgical group was significantly increased( P〈0. 05). Ultrasound imaging analysis showed that model group left ventricular anterior wall thickness of diastole significantly increased 42. 1%,left ventricular posterior diastolic wall thickness significantly increased 31. 1%,left ventricular mass significantly increased 56. 0%( v. s. sham group P〈0. 01). WGA staining showed that myocyte cross-sectional area of surgical group significantly increased( v. s. sham group P〈0. 05). At eight weeks after surgery surgical group left ventricular ejection fraction significantly decreased32. 3%,fractional shortening was significantly decreased 38. 6%( v. s. sham group P〈0. 01). Conclusion:Characteristic of transverse aortic constriction at 4 weeks is left ventricular hypertrophy,simulated the pathological process of cardiac hypertrophy in early heart failure.
