摘 要:目的研究异绿原酸A在大鼠肠肝中的代谢情况。方法建立大鼠原位单向肠-肝血管灌流模型,采用HPLC测定灌流液中异绿原酸A的浓度,计算异绿原酸A在肠、肝的提取率和清除率。结果异绿原酸A的稳态肠提取率和清除率分别为(12.84±1.16)%和(1.28±0.08)m L/min,肝提取率和清除率分别为(68.42±2.20)%和(6.84±0.22)m L/min,肠、肝总提取率和总清除率分别(74.64±4.21)%和(7.46±0.38)m L/min。结论异绿原酸A在肝中有较强的代谢,首过效应明显。Objective To study the metabolism of isochlorogenic acid A in rat intestine-liver. Methods The in situ vascularly perfused rat intestine-liver model was established with a validated HPLC method for assay of the perfusate isochlorogenic acid A,and the extraction ratios and clearance ratios of intestine and liver were investigated. Results The steady state intestinal extraction ratios and clearance ratios in the in situ singlepass perfused rat intestine-liver model for isochlorogenic acid A were( 12. 84 ± 1. 16) % and( 1. 28 ±0. 08) m L / min,respectively. The steady state hepatic extraction ratios and clearance ratios were( 68. 42 ±2. 20) % and( 6. 84 ± 0. 22) m L / min,respectively. The total extraction and clearance ratios were( 74. 64± 4. 21) % and( 7. 46 ± 0. 38) m L / min,respectively. Conclusion The metabolism of isochlorogenic acid A in rat liver was strong,and the first pass effect was strong.
