摘  要:目的:探讨油橄榄叶提取物(olive leaf extract,OLE)和美沙酮对海洛因依赖大鼠肝功能的保护作用.方法:将40只健康大鼠随机分成正常组、OLE组、美沙酮组和OLE+美沙酮组.除正常组外,其余三组动物均按递增法采用皮下注射海洛因建立海洛因依赖大鼠模型,造模后药物处理30d,然后分别用药物进行治疗.用全自动生化分析仪检测血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)和γ-谷酰转肽酶(GGT)活性,用比色法检测大鼠肝脏超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)活性及丙二醛(malondialdehyde,MDA)含量,利用生物显微技术观察肝脏组织结构的变化,采用免疫组织化学法检测肝脏中转化生长因子(transforming growth factor-β1,TGF-β1)和Smad4(drosophila mothers against decapentaplegic 4)的表达情况.结果:OLE+美沙酮组大鼠血清中ALT、AST、GGT的活性明显低于OLE组和美沙酮组;与OLE组和美沙酮组相比,OLE+美沙酮组肝脏SOD、CAT活性明显升高,MDA的含量降低,肝脏中TGF-β1和Smad4的表达明显减弱;光镜下OLE+美沙酮组肝组织病理损害程度较OLE组和美沙酮组明显减轻.结论:OLE和美沙酮联合治疗海洛因造成的肝功能损害疗效优于两种药物的单一用药,两药合用具有保护肝的协同作用.Objective: To explore protection role of olive leaf extract( OLE) and methadone on liver function in heroindependent rats.Methods: Forty rats were randomly divided into control group,OLE group,methadone group and OLE+ methadone group.Each group of rats was subcutaneously given heroin with increasing dosage to establish heroin administration except control group.After modeling,the rats were administrated with OLE and methadone for 30d.The levels of alanine aminotransferase( ALT),aspartate aminotransferase( AST) and γ-glutamyltransferase( GGT) in serum were tested by automatic biochemistry analyzer.Spectrophotometry was used to detect the activities of superoxide dismutase( SOD) and catalase( CAT),and malondialdehyde( MDA) content.The changes of liver histological structure were observed by bio-microscopy.Immunohistochemical analysis was used to determine transforming growth factor-β1( TGF-β1) and Smad4( drosophila mothers against decapentaplegic 4) expression in rat liver.Results: The activities of ALT,AST and GGT in the OLE-treated groups were significant lower in OLE + methadone group than in OLE group and methadone group.Compared with OLE group and methadone group,OLE + methadone group showed that SOD and CAT activities significantly increased,and MDA content and expression of TGF-β1 and Smad4 reduced.The physiological damages in OLE + methadone group were slighter than in OLE group and methadone group.Conclusion:There is better curative effect in OLE combined methadone than in single of OLE or methadone to treat heroin-induced liver function injure,and can enhance liver protection.
