摘 要:目的探索白色念珠菌对罗丹明6G的吸收和泵出的测定意义,并对测定方法进行总结。方法借助2株白色念珠菌敏感株和2株白色念珠菌耐药株,采用多功能微板检测仪、流式细胞仪、荧光显微镜三种仪器,通过定量、半定量和定性的方法测定不同敏感性白色念珠菌对罗丹明6G的吸收和泵出,对定量与半定量结果进行T检验,并与定性结果进行比较。结果不同敏感性白色念珠菌对罗丹明6G的吸收基本一致,实验用耐药菌对罗丹明6G的泵出比敏感菌多;以多功能微板检测仪测定结果为标准,流式细胞仪测定结果与之的一致性为83.3%,荧光显微镜观察结果与之的一致性为91.7%,三种方法具有一致性。结论白色念珠菌对罗丹明6G的吸收和泵出的测定可以为研究新型抗真菌成分的作用机制和真菌耐药逆转剂的作用机制提供工具菌株和可能的研究方向,应根据实验目的和条件选择适当的测定方法。Objective To explore the significance of the absorption and efflux of rhodamine 6G in Candida albicans, and to summarize the testing methods. Methods Multifunctional mieroplate reader, flow cytometry and fluorescence microscope were used to determine the absorption and efflux of rhodamine 6G in Candida albicans with different sensitivity, including 2 sensitive and 2 resistant strains. The resuhs were verified with Students' T test and confirmed by fluorescence microscope. Results The uptake of rhodamine 6G in Candida albicans strains with different sensitivity were nearly the same, while the efflux of rhodamine 6G in resistant strains were more than that of the sensitive ones. The results of flow cytometry and fluorescence microscope were consistent with the data detected by muhifunctional microplate reader ,which was selected as the standard,and the consistencies were 83.3% and 91.7% respectively. Conclusion The detection of uptake and efflux of rhodamine 6G in Candida albicans is of great significance, especially for investigating the mechanisms of antifungal and resistant reversal. An appropriate method should be chosen according to the experiment conditions and purpose.
