摘  要:Rho相关蛋白激酶(ROCK)作为丝氨酸-苏氨酸激酶家族的一个成员是Rho(小GTP酶)第一下游效应器研究表明,诸多心脑血管疾病的发生常伴随着ROCK的异常活化。目前,具有潜在的抑制ROCK活性的小分子化合物已经被开发,并在心脑血管疾病的诊疗中开展了临床测试。本文对ROCK在心脑血管疾病中所发挥的作用及ROCK抑制剂开发应用的最新研究进展进行综述。Rho-assoeiated protein kinase (ROCK) is a member of serine-threonine kinase family that was discovered as a first downstream effector of small GTPase Rho. Some studies have suggested that the abnormal ROCK activation was involved in the patho- genesis of cardiovascular and cerebrovascular diseases. Some small compounds targeting ROCK have been developed and tested in car- diovascular and cerebrovascular diseases clinic models. This review is to introduce the recent advances in the role of ROCK in car- dio-cerebrovascular disease and the application of ROCK inhibitors.
