摘 要:目的:研究分析转化生长因子β1(TGF-β1)基因转染修饰人退变腰椎间盘髓核细胞。方法:选取珠海市人民医院2012年10月至2014年10月收治的60例腰椎间盘退变患者,将其随机分为观察组和对照组,每组30例,观察组使用含有TGF-β1基因的腺病毒载体转染修饰细胞,对照组用EGFP基因(增荧光绿色蛋白基因)转染细胞。观察比较两组相应的编码蛋白表达情况。结果:观察组患者细胞中出现了生长因子TGF-β1,转染率约为30%,对照组没有出现生长因子,但是外源基因均转染成功,有绿色荧光表达,且持续了一个月。结论:使用腺病毒载体能将TGF-β1基因转染患者退变的腰椎间盘髓核细胞,使之产生生长因子TGF-β1,这为腰椎间盘退变的基因治疗提供了基础。Objective To study the TGF- beta 1 gene transfection modified degenerative lumbar nucleus pulposus cells. Methods 60 cases of patients from October 2012 to October 2014 were chosen and randomly divided into observation group and control group, 30 cases in each group and observation group use of adenovirus vector containing the TGF- beta 1 gene transfection modified cells, the control group with EGFP gene(add green fluorescent protein gene transfection cells. Compare two groups corresponding coding protein expression. Results Observation group patients appeared in the cell growth factor TGF- beta 1, transfection rate at about 30%, the control group did not appear growth factor, but the exogenous gene transfection, successfully with green fluorescence expression, and last for a month. Conclusion The use of adenovirus vector can make TGF- beta 1 gene transfection patients with degenerative lumbar nucleus pulposus cells, and make TGF- to generate growth factor beta 1, which provides a basis for gene therapy of lumbar intervertebral degeneration.
