摘  要:目的:探讨表面活性蛋白D基因外显子区域单核苷酸多态性及其蛋白水平与成年女性尿路感染易感性的关系。方法以2011年3月至2013年4月于本院治疗1年以上的尿路感染反复发作的女性患者35例为研究组,同时选取年龄、居住环境、教育水平相似的健康女性31例为对照组。采用聚合酶链反应-序列特异性引物(PCR-SSP)检测两组人群的SP-D-Thr11Met、SP-D-Thr160Ala外显子位点的基因多态性,采用酶联免疫吸附法(ELISA)检测两组人群的血清SP-D蛋白水平。结果 SP-D-Thr11Met基因型分布显示,研究组(n=35)Met/Met、Thr/Met、Thr/Thr基因型分别为8例(22.86%)、20例(57.14%)、7例(20.0%),对照组(n=31)Met/Met、Thr/Met、Thr/Thr基因型分别为7例(22.58%)、18例(58.06%)、6例(19.35%),组间比较差异无统计学意义(P〉0.05)。研究组11Thr、11Met等位基因频率分别为48.57%(34/70)、51.43%(36/70),对照组11Thr、11Met等位基因频率分别为48.39%(30/62)、51.61%(32/62),组间比较差异无统计学意义(P〉0.05)。SP-D-Thr160Ala基因型分布显示,研究组(n=35)Thr/Thr、Thr/Ala、Ala/Ala基因型分别为4例(11.43%)、14例(40.0%)、17例(48.57%),对照组(n=31) Thr/Thr、Thr/Ala、Ala/Ala基因型分别为4例(12.90%)、12例(38.71%)、15例(48.39%),组间比较差异无统计学意义(P〉0.05)。研究组160Thr、160Ala等位基因频率分别为31.43%(22/70)、68.57%(48/70),对照组160Thr、160Ala等位基因频率分别为32.26%(20/62)、67.74%(42/62),组间比较差异无统计学意义(P〉0.05)。研究组血清SP-D蛋白水平为(141.36±86.94)nmol/L,对照组为(62.49±24.37)nmol/L,组间比较差异有统计学意义(t=4.880,P〈0.01)。结论 SP-D外显子区域Thr11Met、Thr160Ala基因变异与成年女性尿路感染易感性无关,但成年女性尿路感染易感患者血SP-D蛋白水�Objective To investigate the correlation between single nucleotide polymorphism (SNP)and protein level of surfactant protein D(SP-D)gene exon locus and susceptibility of urinary tract infection in adult females. Methods A total of 35 female patients with recurrence of urinary tract infection, who were treated in our hospital for more than 1 year between March 2011 and April 2013,were included in the study group,and 31 healthy females with the similar age,living environment and education level were included in the control group. Polymerase chain reaction with sequence specific primers (PCR-SSP) was used to determine the gene polymorphism of SP-D-Thr11Met and SP-D-Thr160Ala exon locus in the two groups. Enzyme linked immunosorbent assay(ELLSA)was used to test the SP-D protein in the two groups.无nbsp;Results The SP-D-Thr11Met genotype distribution showed that the number of Met/Met ,Thr/Met and Thr/Thr genotype were 8 cases(22.86%),20 cases(57.14%)and 7 cases(20.0%)in the study group(n=35),and 7 cases(22.58%),18 cases(58.06%)and 6 cases(19.35%)in the control group(n=31), respectively,without statistically significant differences between the two groups (P〉0.05). The 11Thr and 11Met allele frequency was 48.57%(34/70)and 51.43%(36/70)in the study group,and 48.39%(30/62) and 51.61%(32/62)in the control group,respectively,without statistically significant differences between the two groups(P〉0.05). The SP-D-Thr160Ala genotype distribution showed that the number of Thr/Thr, Thr/Ala and Ala/Ala genotype were 4 cases(11.43%),14 cases(40.0%)and 17 cases(48.57%)in the study group(n=35),and 4 cases(12.90%),12 cases(38.71%)and 15 cases(48.39%)in the control group(n=31),respectively,without statistically significant differences between the two groups(P〉0.05).
