摘 要:目的:探究兔角膜成纤维细胞的原代培养及鉴定方法。方法:选用试验大白兔采集兔角膜成纤维细胞并进行原代培养,应用波形蛋白抗体、角蛋白抗体对体外培养兔角膜成纤维细胞第2代细胞做波形蛋白鉴定。结果:原代培养1∽3 d后可传第1代,1周左右汇合,细胞呈梭形;培养波形蛋白抗体染色阳性、角蛋白抗体染色阴性,鉴定结果显示兔角膜成纤维细胞培养成功。结论:兔角膜成纤维细胞原代培养可行性强、成功率高,可获得状态良好的连续传代扩增细胞,有效支持体外实验研究。Objective To explore the rabbit corneal fibroblasts primary culture and identification methods and results. Methods Experimental rabbits were collected corneal fibroblasts and primary culture, the application of vimentin, cytokeratin antibodies on cultured rabbit corneal fi broblasts second generation cells do vimentin identifi cation. Results The primary cultured 1-3d can pass the first generation, a week or so confluence, cells were spindle; culture vimentin antibody staining, cytokeratin antibody staining was negative, identification showed corneal fibroblasts success. Conclusion The rabbit corneal fibroblasts primary culture feasibility strong, high success rate, can be obtained in good condition serially passaged expanded cells, cultured fi broblasts in vitro and can effectively support the clinical diagnosis and treatment, should be vigorously promote the application.
