摘 要:目的:测定日本蛇根草不同组织器官及发育周期中绿原酸的含量。方法:建立高效液相色谱法。色谱柱ODS Phenomenex Gemini C18柱(250 mm×4.6 mm,5μm),流动相乙腈:0.4%磷酸溶液(1∶9),流速:1.0 ml·min-1,检测波长:327 nm;柱温:30℃。结果:绿原酸在0.022~1.320μg(r=0.999 9)范围内线性关系良好。2月花期采收的日本蛇根草根、茎、叶、花中绿原酸的含量分别为7.076 7,1.030 7,10.609 5,24.110 3 mg·g-1,其全草与四月果期的全草绿原酸含量分别为18.600 4,14.082 6 mg·g-1。结论:日本蛇根草不同组织器官绿原酸含量差异较大,其中花的含量最高。不同发育周期绿原酸的含量也不同,花期含量明显高于果期。Objective: To determine the content of chlorogenic acid in different tissues at different cyclogeny of Ophiorrhiza japoni- ca BI. Methods: An HPLC method was established with an ODS Phenomenex Gemini Cls column (250 mm x 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile: 0.4% phosphoric acid solution ( 1 : 9), the flow rate was 1.0 ml · min - 1, the detection wave- length was at 327 nm and the column temperature was 30 ℃. Results: There was a good linear relationship of chlorogenic acid within the range of 0. 022-1. 320 μg( r = 0.999 9). The content of chlorogenic acid in roots, stems, leaves and flowers of Japanese snakeroot harvested in February flowering period was 7. 076 7, 1. 030 7, 10.609 5and 24. 110 3 mg · g-1, respectively. Meanwhile, that was 18. 600 4 mg· g-I and 14.082 6 mg · g-1 in all grass harvested in February and April. Conclusion: The content of chlorogenic acid in different tissues of Ophiorrhiza japonica BI. in different growth stages is various. The flowers have the highest content, and the con- tent in flowering period is significantly higher than that in fruit period.
