摘 要:目的探讨中药活性成分对胶原诱导的关节炎(collagenⅡ-induced arthritis,CIA)鼠关节滑膜细胞增殖的影响,为自身免疫性关节炎的治疗提供一定的依据。方法采用MTT法检测不同中药活性成份(雷公藤甲素、瑞香素、紫杉醇、紫草素和槲皮素)对CIA鼠关节滑膜细胞增殖的影响。选用抑制效果好的中药活性成份进一步探讨其对关节滑膜细胞的作用机制,并采用Western Blot检测中药活性成份对细胞促凋亡基因Fas L、p53、DR3和PDCD5表达的影响。结果 5种中药活性成分不同浓度时对CIA鼠关节滑膜细胞的增殖均有抑制作用,并均呈现剂量依赖性,其中抑制效果较好的是瑞香素和雷公藤甲素。瑞香素组和雷公藤甲素组DR3、Fas L、p53表达水平均明显高于对照组(P<0.05)。瑞香素组中促凋亡基因中p53的表达明显高于雷公藤甲素组(P<0.05),但2组DR3、PDCD5、Fas L差异无统计学意义(P>0.05)。结论雷公藤甲素、瑞香素、紫杉醇、紫草素和槲皮素对自身免疫性关节炎大鼠关节滑膜细胞的增殖均有抑制作用,并均呈现剂量依赖性,抑制效果较好的是瑞香素和雷公藤甲素。瑞香素和雷公藤甲素均可促进自身免疫性关节炎鼠关节滑膜细胞中促凋亡基因DR3、Fas L和p53的表达,从而促进滑膜细胞凋亡,抑制其增殖。Objective To provide theoretical basis for the drug treatment of autoimmune arthritis by studying the influence of active ingredients extracted from traditional Chinese medicine on the proliferation in the joint synovial cells of collageninduced arthritis (CIA) rats. Methods Effects of different active ingredients of traditional Chinese medicine on synovial cell proliferation of CIA rats were detected by MTT method. The expression of FasL, p53, DR3 and PDCD5 of synovial cells of collagen-induced arthritis rats were detected by Western Blot. Results Five kinds of active ingredients extracted form traditional Chinese medicine had different degrees of inhibition to the synovial cells of autoimmune arthritis rats and presented good dose dependent. And among the 5 kinds of active ingredients, triptolide and daphnetin were better. The expression of DR3, FasL and p53 detected in the joint synovial cells of collagen-induced arthritis rats treated with daphnetin or triptolide was higher than that in untreated CIA joint synovial cells (P 〉0.05). Compared with triptolide treatment group, a higher expression of p53 in CIA joint synovial cells treated with daphnetin was observed. However, there was no significant difference in DR3, PDCD5 and FasL between daphnetin treatment group and triptolide treatment group (P 〉0.05). Conclusion Triptolide, daphnetin, paclitaxel, shikonin and quercetin have different degrees of inhibition to the synovial cells of autoimmune arthritis rats and present good dose dependent. And among the 5 kinds of active ingredients, triptolide and daphnetin are better. Additionally, the expression of pro-apeptetic genes DR3, FasL and p53 is remarkably increased after being administered with daphnetin or triptolide, which could promote cell apoptosis.
